Natural ingredients are gaining increasing attention from manufacturers following consumers’ concerns about the excessive use of synthetic ingredients. However, the use of natural extracts or molecules to achieve desirable qualities throughout the shelf life of foodstuff and, upon consumption, in the relevant biological environment is severely limited by their poor performance, especially with respect to solubility, stability against environmental conditions during product manufacturing, storage, and bioavailability upon consumption. Nanoencapsulation can be seen as an attractive approach with which to overcome these challenges. Among the different nanoencapsulation systems, lipids and biopolymer-based nanocarriers have emerged as the most effective ones because of their intrinsic low toxicity following their formulation with biocompatible and biodegradable materials. The present review aims to provide a survey of the recent advances in nanoscale carriers, formulated with biopolymers or lipids, for the encapsulation of natural compounds and plant extracts.

Prevalensi dan morbiditas penyakit kanker terus meningkat. Pencarian agen sitotoksik baru terus dilakukan termasuk yang berasal dari bahan alam. Pengembangan sistem penghantaran untuk meningkatkan aktivitas sitotoksik banyak dilakukan seperti dengan pembentukan fitosom, salah satu sistem nanovesikular. Tujuan dari penelitian ini adalah untuk mengkaji pengembangan bahan-banan herbal dengan sistem penghantaran fitosom dan juga mengetahui pengaruhnya terhadap aktivitas sitoksik dari senyawa bahan alam tersebut. Penelitian dilakukan dengan melakukan kajian literatur dari artikel yang berasal dari database bereputasi yang memenuhi kriteria inklusi dan eksklusi sesuai tujuan penelitian. Dari hasil kajian diketahui bahwa sistem fitosom sudah banyak dikembangkan untuk penghantaran agen sitoksik herbal seperti pada ekstrak aloe vera, celastrol, curcumin, curcuminoid, diosgenin, icariin, genistein, lycopene, mytomicin, propolis, silibinin, sinigrin, dan thymoquinone. Fitosom diformulasikan dengan menggunakan fosfatidilkolin pada perbandingan fitokonstituen: fosfolipid yakni 1:(1-3), dengan atau tanpa penambahan kolesterol. Pengembangan sediaan fitosom terbukti mampu meningkatkan aktivitas sitotoksik bahan herbal baik berdasarkan pengujian in-vitro atau in-vivo. Pada pengujian in-vitro peningkatan aktivitas ditandai dengan penurunan nilai IC50 dengan tingkat penurunan yang bervariasi dalam rentang 1,7-11 kalinya. Ukuran partikel yang lebih kecil dengan struktur fosfolipid bilayer diketahui menjadi penyebab utama terjadinya peningkatan aktivitas tersebut. Dari hasil kajian disimpulkan bahwa sistem fitosom potensial untuk mampu meningkatkan aktivitas sitotoksik senyawa herbal.

How Should the Worldwide Knowledge of Traditional Cancer Healing Be Integrated with Herbs and Mushrooms into Modern Molecular Pharmacology?

Traditional herbal medicine (THM) is a “core” from which modern medicine has evolved over time. Besides this, one third of people worldwide have no access to modern medicine and rely only on traditional medicine. To date, drugs of plant origin, or their derivates (paclitaxel, vinblastine, vincristine, vinorelbine, etoposide, camptothecin, topotecan, irinotecan, and omacetaxine), are very important in the therapy of malignancies and they are included in most chemotherapeutic regimes. To date, 391,000 plant and 14,000 mushroom species exist. Their medical and biochemical capabilities have not been studied in detail. In this review, we systematized the information about plants and mushrooms, as well as their active compounds with antitumor properties. Plants and mushrooms are divided based on the regions where they are used in ethnomedicine to treat malignancies. The majority of their active compounds with antineoplastic properties and mechanisms of action are described. Furthermore, on the basis of the available information, we divided them into two priority groups for research and for their potential of use in antitumor therapy. As there are many prerequisites and some examples how THM helps and strengthens modern medicine, finally, we discuss the positive points of THM and the management required to transform and integrate THM into the modern medicine practice.
