Evaluation of an Immunoradiometric Assay for Bone Alkaline Phosphatase Mass Concentration in Human Sera

Abstract: The performance characteristics of an immunoradiometric assay for bone alkaline phosphatase mass concentration in human sera are reported.Within-run imprecision (n = 20) was 12.1% (x = 7.8 μ §/\) and 3.6% (x = 22.8 Mg/1), between-day imprecision (n = 8) was 10.1% (X = 20.3 μ£/1) and 2.8% (x = 84.3 μ §/1). There was a linear relationship between the concentrations of the Standards employed and the counts per minute up to 120 μ^Ι. The detection limit was 0.3In 102 apparently healthy persons (51 males and 51 fema… Show more

“…For the interpretation of bone alkaline phosphatase values (as determined immunoradiometrically) it has to be taken into account that the antibodies employed in the test show 16% cross reactivity with liver alkaline phosphatase (19,20). In the sera examined in the present study interference by liver alkaline phosphatase can be excluded since all sera with increased bone alkaline phosphatase mass concentrations showed Ζ,-γ-glutamyl transferase activity concentrations within the reference range.…”

“…Both assays show a cross-reactivity of 14-20% with liver alkaline phosphatase (8)(9)(10)(11)(12). However, it is supposed that in patients with hepatobiliary diseases the measured bone alkaline phosphatase values stay within the reference range if total alkaline phosphatase activity does not exceed about twice the upper limit of the normal range (6).…”
