Background: Pneumatic pulsation therapy may combine the effects of cupping therapy and massage. This study investigated the effect of pneumatic pulsation therapy on chronic neck pain compared to standard medical care. Methods: 50 patients (79.15% female; 46.17 ± 12.21 years) with chronic nonspecific neck pain were randomized to treatment group (TG; n = 25) or control group (CG; n = 25). The TG received 5 pneumatic pulsation treatments over a period of 2 weeks utilizing a mechanical device. Treatment was applied as a combination of moving and stationary pulsating cupping. Main outcome measure was pain intensity in pain diaries (numerical rating scale). Secondary outcome measures included functional disability (NDI), quality of life (SF-36), and pain at motion. Sensory thresholds, including pressure pain threshold, were measured at pain-related sites. Results: After the intervention, significant group differences occurred regarding pain intensity (baseline: 4.12 ± 1.45 in TG and 4.20 ± 1.57 in CG; post-intervention: 2.72 ± 1.62 in TG and 4.44 ± 1.96 in CG; analysis of covariance: p = 0.001), NDI (baseline: 25.92 ± 8.23 and 29.83; post-intervention: 20.44 ± 10.17 and 28.83; p = 0.025), and physical quality of life (baseline: 43.85 ± 7.65 and 41.66 ± 7.09; post-intervention: 47.60 ± 7.93 and 40.49 ± 8.03; p = 0.002). Further significant group differences were found for pain at motion (p = 0.004) and pressure pain threshold (p = 0.002). No serious adverse events were reported. Conclusion: Pneumatic pulsation therapy appears to be a safe and effective method to relieve pain and to improve function and quality of life in patients with chronic neck pain.

(2015) Hypolipidemic effects of Solidagochilensis hydroalcoholic extract and its major isolated constituent quercetrin in cholesterol-fed rats, Pharmaceutical Biology, 53:10, 1488-1495, DOI: 10.3109/13880209.2014 Objective: This study evaluates the hypolipidemic and antioxidant effects of hydroalcoholic extract (HE) and quercetrin from S. chilensis aerial parts in cholesterol-fed rats. Materials and methods: The HE was analyzed by high-performance liquid chromatography, followed by quercetrin isolation. Hypercholesterolemic rats (1% cholesterol and 0.5% cholic acid for 15 d) were treated with HE (150, 300, and 600 mg/kg p.o.; n ¼ 6), simvastatin (4 mg/kg p.o.; n ¼ 6), or quercetrin (10 mg/kg p.o.; n ¼ 6) once a day for 30 d. During this period, a highcholesterol diet was maintained until the 30th day of treatment.Results: Rats treated with HE (150, 300, and 600 mg/kg) and quercetrin showed decreased serum levels of total cholesterol (À19.9, À27.5, À31.0, and À39.4%), lipoprotein-cholesterol (À36.0, À37.5, À43.3, and À59.4%), and triacylglycerides (À15.6, À23.5, À29.8, and À27.2%) when compared with the control group similar to simvastatin. Moreover, treatment with HE and quercetrin decreased hepatic 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase activity (35.1% on average) and increased fecal cholesterol levels (38.2% on average). Discussion and conclusions: Our results suggest that hypolipidemic effects of HE are associated with it modulating the activity of HMG-CoA reductase and its interference in the reabsorption and/or excretion of intestinal lipids. Solidago chilensis and its main constituent, quercetrin, may thus be effective as cholesterol-lowering agents and in preventing atherosclerosis.

RESUMO:A espécie Solidago chilensis Meyen, Asteraceae é conhecida como erva-lanceta ou arnica-brasileira, sendo utilizada popularmente como antimicrobiana e para o tratamento de inflamações tópicas. No entanto, estudos fitoquímicos e farmacológicos para as partes aéreas são escassos. Neste trabalho, realizou-se a determinação de flavonoides por espectrofotometria de UV/Vis, prospecção fitoquímica da fração acetato de etila visando o isolamento do constituinte químico majoritário e validação analítica por cromatografia líquida de alta eficiência (CLAE). O teor de flavonoides totais foi de 5,42%, representados como hiperosídeo. O fracionamento químico utilizando métodos cromatográficos (cromatografia líquida em coluna gel de sílica; CHCl 3 :EtOH; 8:2 v/v) e espectroscópicos ( 1 H RMN, 13 C RMN e ESI-MS) revelou o isolamento de quercetina-3-O-α-L-ramnosídeo(quercitrina). A sensibilidade e a linearidade (r = 0,999) da validação analítica, utilizando a quercitrina isolada do extrato hidroalcoólico da planta, revelaram um rendimento de 5,29% do analito em relação à droga vegetal. Precisão, recuperação e robustez, além dos valores estabelecidos para os limites de detecção (LOD) e de quantificação (LOQ), poderão ser utilizados como parâmetros de qualidade para extratos à base de S. chilensis. ABSTRACT: Chemical isolation and analytical validation by high-performance liquid chromatography of quercitrin in Solidago chilensis Meyen (Asteraceae).The speciesSolidago chilensis Meyen Asteraceae, known as "erva-lanceta" or "Brazilian arnica", is popularly used as an antimicrobial and topical treatment for inflammations. However, phytochemical and pharmacological studies of its aerial parts are scarce. In this study, flavonoids were determined by UV/Vis spectrophotometry and phytochemical screening of the ethyl acetate fraction with the goal of isolating the major chemical constituent and analytically validating it through high performance liquid chromatography (HPLC). The total flavonoid content was 5.42%, represented as hyperoside. Chemical fractionation using chromatographic (liquid chromatography in column of silica gel, CHCl3:EtOH, 8:2 v/v) and spectroscopic methods (1H RMN, 13C RMN, and ESI-MS) revealed the isolation of quercetin-3-O-α-L-rhamnoside (quercitrin). The sensitivity and linearity (r = 0.999) using the isolated quercitrin of the hydroalcoholic extract of the plant revealed a yield of 5.29% of analyte in relation to the plant. Precision, recovery, and robustness, as well as values set for the limits of detection (LOD) and quantitation (LOQ) can be used as quality parameters for extracts based on S. chilensis.
