Evaluation of Allergenicity of Egg Yolk Immunoglobulin Y and Other Egg Proteins by Passive Cutaneous Anaphylaxis

Abstract: The allergenicity of egg yolk immunoglob ulin (IgY) was studied using passive cutaneous anaphy laxis. It was observed that the intact yolk, water soluble fraction of the yolk, whole IgY molecule or its antigen binding (Fab9 ) and crystallizable (pFc9 ) fragments produced by pepsin digestion, elicited low IgE antibody response in mice when com pared to egg white, a known allergen. The pFc9 fragment of IgY was more antigenic than the Fab9 fragment as determined by ELISA. Low cross-reactivity was also observed be… Show more

“…As a result, dietary antigens are presented in association with signals that drive a suppressive immune response. It has been reported that IgY is highly immunogenic as it has been used as a model antigen for testing mucosal adjuvants, which might explain the presence of IgG anti IgY in sera of treated animals [69], [70]. On the contrary, the oral administration of VHH nanoAbs might induce a tolerant state, preventing the host from eliciting an anti-VHH immune response, but further studies must be conducted to confirm this statement.…”

“…In addition, it has been demonstrated in pre-clinical studies that eggs of hyperimmunized hens possessed anti-inflammatory and anti-diarrheic properties and offer benefits to a number of different human structures and functions including the circulatory and immune systems and the joints [35] . Akita and co-workers showed that egg yolk as well as purified IgY Abs were immunogenic but failed to induce a detectable IgE Ab response in intraperitoneally immunized mice [36] . In this study we evaluated the protection against HRV diarrhea conferred by the prophylactic administration of HRV specific IgY Abs to gnotobiotic piglets experimentally inoculated with the prevalent strain of HRV, Wa G1P[8].…”

“…The enzymatic digestion even promoted the release of immunoreactive peptides with IgE‐binding epitopes of fragment 57–83 and fragment 108–122 that are linked by disulfide bonds. In the work done by Akita et al ., the allergenicity of egg yolk IgY was studied using passive cutaneous anaphylaxis (PCA) test and enzyme‐linked immunosorbent assay. It was observed that the hydrolysate fragments produced by pepsin digestion exhibited lower IgE antibody response, which was associated with higher suppressor T‐lymphocytes or lower helper T‐lymphocytes, or both.…”
