Evaluation of agomelatine stability under different stress conditions using an HPLC method with fluorescence detection: application to the analysis of tablets and human plasma

Abstract: A simple and highly sensitive stability-indicating HPLC method was developed and validated for the determination of the new antidepressant agent, agomelatine (AGM). Separation of AGM from its stress-induced degradation products was achieved on a BDS Hypersil phenyl column (250 mm × 4.6 mm i.d., 5 µm particle size) using methanol-0.05 M phosphate buffer of pH 2.5 (35: 65, v/v) as a mobile phase with fluorescence detection at 230/370 nm. Naproxen was used as an internal standard. The method satisfied all the val… Show more

“…[2] It has proven to have anxiolytic properties and thus may prove to be very useful in the treatment of anxiety disorders. [3] The literature reveals that very few analytical methods have reported the estimation of AGM in different matrices including spectrophotometry [4] , high pressure liquid chromatography (HPLC) [4][5][6][7][8][9][10] and LC-MS. [11] None of the literature review indicates spectrofluorimetric methods for AGM. Therefore, an attempt has been made to develop spectrofluorimetric methods for AGM.…”

“…It is a selective agonist of the human melatonergic MT 1 and MT 2 receptors and also shows 5-HT 2C receptor antagonist activity. [3][4] Very few analytical methods have been reported for the determination of AGM such as HPLC, [5][6][7][8][9][10][11] LC-MS/MS in human plasma and HPTLC. 14 So, at present the authors have developed a stability indicating RP-HPLC method for the determination of AGM in presence of its degradation products.…”

“…A stability study ensures the maintenance of product quality, safety, and efficacy throughout its shelf life. Stress testing can help identify degradation products and provide important information about the intrinsic stability of drug substances [ 9 , 10 ]. Regulatory agencies recommend the use of stability-indicating assay methods for the analysis of stability samples [ 11 ].…”
