Evaluation of a water-soluble adjuvant for the development of monoclonal antibodies against small-molecule compounds

Abstract: A water-soluble adjuvant named QuickAntibody (QA) was introduced into the procedure of mouse immunization for the development of hapten-specific monoclonal antibodies (mAbs), using four kinds of pesticides as model compounds. Compared with conventional Freund's adjuvants, QA treatments offered relatively low but acceptable antiserum titers after three inoculations, gave little adverse effects to the experimental animals, and were preferable in harvesting splenocytes during the steps of cell fusion. Afterwards,… Show more

“…The antisera (TPAbs-1−6) exhibited different affinities to the coating antigen (T-OVA) with titers ranging from 64 000 to 256 000, which were higher than those of previously reported antisera against various pesticides. 48 TPAbs-4, which had the highest titer (256 000), was selected for antibody purification. The concentration of the purified polyclonal antibody was about 4.52 mg/mL.…”

“…Thiacloprid-C4C4 mAb was prepared according to the protocol described in our previous work [ 47 ]. The thiacloprid–bull serum albumin (BSA) conjugate reported by Zhenjiang Liu et al [ 32 ] was used as the artificial immunogen to induce the immune response.…”

“…Enzyme-linked immunosorbent assay (ELISA) was used to confirm the conjugates by the standard procedure (Liu R et al, 2016;Song et al, 2017). OTA-BSA (OTA-OVA) or ZEN-BSA (ZEN-OVA) was immobilized on polystyrene 96-well plates and then the coated antigens were probed with their specific monoclonal antibody, followed by the addition of goat anti-rabbit IgG-horseradish peroxidase (HRP) to each well.…”
