Evaluation of a microplate spectrophotometer for soil organic carbon determination in south‐central Idaho

Abstract: Determination of soil organic carbon (SOC) is highly desirable for assessing fertility and carbon sequestration; however, numerous methods of determination warrant study of method agreement. Recently, a novel method was developed following dichromate oxidation using a microplate spectrophotometer. This novel method was compared with (i) total C by dry combustion-soil inorganic carbon (DC w/o pretreatment-Pcal); (ii) traditional Walkley-Black titration (WBTIT) and (iii) loss on ignition (LOI 360°C) in calcareou… Show more

“…The NO 3 − ‐N was determined by extracting the soil with calcium chloride and measuring the N concentration in the extract in a flow injection analyzer (FIA) (Houba et al., 2000). The SOM was determined using the loss‐on‐ignition method (Bierer et al., 2021; Nelson & Sommers, 1996; Schulte & Hopkins, 1996). The SOM determination was based on gravimetric weight loss caused by the oxidation of SOM in a high‐temperature reaction.…”

“…The alkali‐hydrolyzable nitrogen (AN) and available phosphorous (AP) were determined by the alkali‐hydrolyzed reduction diffusing method and colorimetric assay method, respectively (Yu et al, 2022). Soil organic carbon (SOC) was measured by the potassium dichromate oxidation‐spectrophotometric method (Bierer et al, 2021). Soil total carbon (TC) and dissolved organic carbon (DOC) were quantified on a CHNOS elemental analyzer (Vario EL, Germany) and TOC analyzer (Shimadzu TOC‐VCPH, Japan), respectively.…”

“…Organic carbon was determined by the Walkley-Black method ( Bierer et al, 2020 ). The percentage of organic matter was used to calculate the organic carbon content by using the conversion factor 1.724 and the fact that 58% of the soil organic matter is the average content of carbon.…”
