Evaluation of a Competitive ELISA for Detection of Antibodies against Avian Influenza Virus Nucleoprotein

Abstract: A competitive enzyme-linked immunosorbent assay (C-ELISA) employing a baculovirus-expressed recombinant nucleoprotein and a monoclonal antibody was developed for the detection of antibodies to type A influenza virus nucleoprotein. The performance of the C-ELISA was evaluated by testing 756 chickens, 1123 turkeys, 707 emus, and 1261 ostriches, for a total of 3847 serum samples. Relative to the agar gel immunodiffusion (AGID) test, the C-ELISA had a sensitivity of 100% for all four species. The C-ELISA's sensiti… Show more

“…Serologic evidence of influenza A was detected by using a competitive ELISA (C-ELISA) specific to influenza A nucleoprotein (NP) as described by Zhou et al (1998). Briefly, microtiter plates coated with recombinant NP were incubated with a 1:5 dilution of each serum sample immediately followed by addition of the monoclonal antibody to NP.…”

“…Infl uenza A virus specifi c antibodies in serum were detected using a competitive ELISA (cELISA) specifi c for the infl uenza A virus nucleoprotein (NP) [23] as well as IDEXX HerdChek SIV Test Kit (IDEXX Laboratories, Westbrook ME). A virus neutralization test (VNT) was conducted in 96-well microtiter plates (Corning Incorporated, NY), according to methods described in "WHO Manual on Animal Infl uenza Surveillance and Diagnosis" [24].…”
