Fasciola hepatica is a parasite that is located in the liver of ruminants with the possibility to infect horses, pigs and humans. The parasite belongs to the Trematoda class, and it is the agent causing the disease called fasciolosis. This disease occurs mainly in temperate regions where climate favors the development of the organism. These conditions must facilitate the development of the intermediate host, the snail of the genus Lymnaea. The infection in domestic animals can lead to decrease in production and control is made by using triclabendazole. Triclabendazole resistance in F. hepatica has been reported worldwide including in Brazil. Another concern is the increase number of human cases with the consumption of contaminated vegetables in regions where sanitation is inadequate together with the presence of infected animals and the absence of efficient control methods. The knowledge of the epidemiology of animal fasciolosis, including their occurrence, distribution and monitoring with techniques such as PCR and ELISA is reaching a new level with the usage of the Geographic Information System. The objective is to use new technologies for early fasciolosis diagnostic, as well as, to develop geoprocessing technics that could allow the determination of its prevalence and the evolution of clinical cases in animals before hand. This review paper provides an overview of F. hepatica, covering the aspects listed above, including original data. Key words: Fasciola hepatica, epidemiology, geographic information system, zoonosis ResumoFasciola hepatica é um parasita que se localiza no fígado de ruminantes, podendo infectar equinos, suínos e humanos. O parasito pertence à classe Trematoda, sendo o agente responsável pela doença denominada fasciolose, que ocorre principalmente em regiões temperadas, com clima favorável para o desenvolvimento do organismo. Tais condições devem auxiliar no desenvolvimento do hospedeiro intermediário, o molusco do gênero Lymnaea. Existe uma crescente preocupação quanto ao número de casos humanos diagnosticados em certas regiões cujo saneamento básico é inadequado, há presença de animais infectados e cultura em consumir hortaliças de áreas contaminadas cruas ou pouco cozidas. O parasitismo em animais domésticos pode levar a uma queda no desempenho animal e seu controle é realizado, principalmente, com o uso do triclabendazole. Entretanto, relatos quanto à resistência do patógeno à este produto vêm sendo descritos, inclusive no Brasil. O conhecimento da epidemiologia da fasciolose animal, incluindo sua ocorrência, distribuição e monitoramento com técnicas laboratoriais como o ELISA e a PCR, estão tendo um novo progresso com a utilização do Sistema de Informação Geográfica. O objetivo de utilizar essas novas tecnologias é diagnosticar a fasciolose mais precocemente, assim como, desenvolver técnicas de geoprocessamento que possam determinar a prevalência e a evolução dos casos em animais. In the present study, the carbohydrate structures associated with Fasciola gigantica adult worm were identified by indirect hemagglutination inhibition test. Glucose was found to be the main monosaccharide associated with the fluke. According to indirect hemagglutination inhibition results, purification of glycoprotein fractions from worm crude extract was carried out by affinity chromatography immobilized glucose agarose gel and Con-A lectin columns. The isolated glycoprotein fractions, FI and FII, were characterized by SDS-PAGE which revealed one band in FI of 26 kDa and another one band of 19.5 kDa in FII compared with 12 bands associated with whole worm extract. Both fractions were also characterized by isoelectric focusing technique which proved that both bands were acidic in nature with pIs 6.4 and 6.5 respectively. The comparative diagnostic evaluation of the two isolated glycoprotein fractions and crude extract of experimental fasciolosis in rabbits by ELISA revealed that FII was more potent in the diagnosis during prepatent (first week post infection) and patent periods (10 weeks post infection) than FI and crude extract. Moreover, infected rabbit sera at ten weeks post infection identified both bands; 26 and 19.5 kDa in western blot analysis confirming its immunodiagnostic activities which was proved previously by ELISA. FII proved potency in diagnosis of fasciolosis in 200 buffalo serum samples of different ages and sexes using ELISA which recorded 95 % positive and 5 % negative samples. Moreover, the detailed structural analyses of the most potent fraction, F11, using mass spectrum was made and elucidated chemical structure; O-glycan [Ser-(Arg-Ser-Arg-Ser-GlucNAc) 19 -GlucNAc]. The present result introduces GlucNAc rich fraction of F .gigantica that can be used successfully in the diagnosis of acute and chronic fasciolosis.

Un sondage sérologique sur la fasciolose a été réalisé en Guinée sur des sérums issus de la sérothèque du Laboratoire de diagnostic vétérinaire de la Direction nationale des services vétérinaires. Au total, 480 sérums provenant de bovins N’Dama ont été sélectionnés par randomisation et analysés par la technique de dosage immuno-enzymatique (Elisa). Les résultats obtenus ont mis en évidence la présence d’anticorps de douves chez 44,1 p. 100 des bovins, avec un taux de 35,5 p. 100 dans la région Ouest de la Guinée et de 55,5 p. 100 dans la région Est. Les bovins de tous les âges ont été séropositifs avec des taux variant de 27,8 à 44,5 p. 100.
