Evaluating the Effect of Dye–Dye Interactions of Xanthene-Based Fluorophores in the Fluorosequencing of Peptides

Abstract: A peptide sequencing scheme utilizing fluorescence microscopy and Edman degradation to determine the amino acid position in fluorophorelabeled peptides was recently reported, referred to as fluorosequencing. It was observed that multiple fluorophores covalently linked to a peptide scaffold resulted in a decrease in the anticipated fluorescence output and worsened the single-molecule fluorescence analysis. In this study, we report an improvement in the photophysical properties of fluorophore-labeled peptides by… Show more

“…Previously, we found that attaching fluorophores to peptides using flexible (PEG) 10 linkers provided a modest reduction of dye-dye quenching (Bachman et al 2022). However, we suspected that the short length (approx.…”

“…Since xanthene-like derivatives serve as biomarkers in various biological assays using the donor–acceptor model and also have a potential role in neurodegenerative disease, we decided to build a library (Figure ) using it as a fundamental framework. In the neuro-regeneration study, any probe displaying hMSCs trans-differentiation to neurons is unknown.…”
