A technique for embryonic implantation and the subsequent recording of electrocortical, neck muscle, and ocular activity continously from the 20th day of incubation through hatching and the first few days thereafter is demonstrated. The embryonic maturation of the EEG, with a characteristic muscle burst pattern heralding hatching was found, supporting previous reports obtained with acute preparations. The technique for injection into the chorioallantoic membrane (CAM) vessels or direct deposition onto the CAM is also described. The usefulness of the embryonic neurophysiological implantation coupled with the injection at specific stages of development is discusses as an approach to the understanding of the parameters of the maturation of the sleep-wakefulness cycle, neurochemistry, and behavior.

The organization of sleep and wakefulness after maternal separation in young rats

Two-week-old infant rats were studied before and after 24 hr of maternal absence. This experience produced an increase in time spent awake, a reduction in time spent in paradoxical sleep (PS), and an increased frequency of state transitions with more frequent and shorter periods of both slow-wave sleep (SWS) AND PS. Body movements were increased and nonnutritive sucking decreased during PS after separation. Other aspects of sleep organization were relativly stable: the pattern of probability of direction of state transitions, the integration of the physiological and behavioral determinants of state, the pattern and amount of nonnutritive sucking, and the underlying PS cycle period. These findings, and the results of 2 control experiments, suggest that some aspects of the prolonged absence of the mother produce a shift in the infant's central neutral state regulation through processes yet to be understood.

Control of sleep‐wake states in the infant rat by features of the mother‐infant relationship

A series of experiments in 2 strains of Wistar rats explored which aspects of maternal separation were responsible for the sleep disturbance of the young. Neither their own mother, after mammary duct ligation, nor constant intragastric infusion of nutrient during the day of separation were found to ameliorate significantly the changes in sleep-wake state organization. However, a periodic nutrient delivery schedule and a formula closely resembling rat milk each ameliorated, and together entirely prevented, the sleep-wake disturbance. Decreased ambient temperature during separation tended to exaggerate the disturbance and periodic tactile-vestibular stimulation tended to reduce it. These findings suggest that the infant rat's normal sleep-wake pattern is maintained by the rhythmicity and composition of the milk delivered to it by its mother, although her warmth and behavioral interaction with it may play subsidiary roles. Separation produces its effects by removing these regulating features of the relationship.
