Ethylene Does Not Promote Adventitious Root Initiation on Apple Microcuttings

Abstract: We investigated the role of ethylene on adventitious rooting of `Gala' (easy-to-root) and `Triple Red Delicious' (difficult-to-root) apple (Malus domestica Borkh.) microcuttings. Root count increased significantly as IBA level increased, with highest root counts on `Gala'. Ethylene evolution increased significantly with IBA level without significant differences between cultivars. Basal section removal of microcuttings in the area of root origin reduced root count without chan… Show more

“…It is difficult to directly compare the results of Harbage and Stimart (1996) with our own. Although there was no effect of ethylene on apple rooting, they reported only root number with no information on rooting percentage, root elongation or callus formation.…”

“…In this study, we tested the effects of ACC, AgNO 3 , AVG and CoCl 2 on root regeneration from shoot cultures of the apple cultivar Royal Gala, for which we have developed a transformation system (Yao et al 1995). After these experiments were completed, Harbage and Stimart (1996) published results suggesting that ethylene was not associated with the rooting of apple microcuttings.…”

“…Ethylene response has been shown very specifically, and conflicting responses have been found from the same species (Harbage and Stimart 1996;Ma et al 1998). Hence it follows that the effects of ethylene vary markedly between species or even within the same species.…”

“…We hypothesized that ethephon exerts its effect on increasing shoot formation in 'Little Pinkie' through its decomposition to ethylene in the headspace of culture vessels, and thus investigated the putative effects of ethephon and ethylene on the plants in a culture vessel capable of gas exchange. Previous studies on the presence of ethylene in vitro have merely described the negative/positive effects of ethylene/ethephon on growth variables of plants (Harbage and Stimart, 1996;Kerbauy and Colli, 1997), in contrast to detailing an ethephon-ethylene relationship within an unsealed culture vessel used for in vitro culture. The culture vessels used in our study needed to be unsealed to allow for gas exchange during plant growth in vitro.…”
