Seeds of Nigella sativa (black cumin/kalonji) used in pickles as spice, have also been traditionally used in treatment of many diseases including diabetes and hypertension. Among many activities exhibited by N. sativa and its constituents in animal experiments, antidiabetic property is most important. Thymoquinone (TQ), a volatile oil, is one of its active constituents but antidiabetic activity has also been shown by its aqueous extract and defatted extract. N. sativa may be beneficial in diabetic individuals and those with glucose intolerance as it reduces appetite, glucose absorption in intestine, hepatic gluconeogenesis, blood glucose level, cholesterol, triglycerides, body weight and simulates glucose induced secretion of insulin from beta-cells in pancreas; improves glucose tolerance as efficiently as metformin; yet it has not shown significant adverse effects and has very low toxicity. In streptozotocin (STZ) induced diabetic rats it causes gradual partial regeneration of pancreatic beta-cells, increases the lowered serum insulin concentrations and decreases the elevated serum glucose. N. sativa has antioxidant activity and protective role of TQ against development of type I diabetes may be via NO inhibitory pathway. It also exerts an insulin-sensitizing action in hepatocytes. Seeds of N. sativa have been safely consumed by human patients in many clinical trials which however were not aimed to assess its antidiabetic activity. In future clinical studies may show potential of N. sativa, its constituents or their synthetic analogues, in prevention and control of diabetes.

Context: Cactus pear (Opuntia ficus-indica (L.) Mill. (Cactaceae)) is a medicinal plant widely used to treat diabetes. Objective: This work investigates the hypoglycemic and antihyperglycemic effect of cactus pear seed oil (CPSO), its mechanism of action, and any toxic effects. Materials and methods: The hypoglycemic effect of CPSO was evaluated in groups of six healthy Wistar rats given 1 or 2 ml kg À1 orally and compared with groups receiving glibenclamide (2 mg kg À1 ) or water. Glycemia was determined after 30, 60, 120, 240, and 360 min. The antihyperglycemic effect of CPSO was determined in healthy rats and in streptozotocin-induced diabetic rats (STZ); normal rats received 0.8 ml kg À1 CPSO, while diabetic rats received 1 ml kg À1 CPSO, their controls received water or 2 mg kg À1 glibenclamide. For the antihyperglycemic effect evaluation, all the animals were fasted for 16 h before treatment and received glucose orally at 1 g kg À1 30 min after treatment; blood was taken after 30, 90, 150, and 210 min. Intestinal glucose absorption was estimated in rat jejunum perfused with a solution containing 5.55 mmol l À1 glucose. Acute toxicity was determined in albino mice that received oral or intraperitoneal doses of 1, 3, or 5 ml kg À1 CPSO. Results: CPSO (p.o.) decreased postprandial hyperglycemia (60 min after glucose loading), 40.33% and 16.01%, in healthy and STZ-diabetic glucose-loaded rats, respectively. CPSO, also, significantly decreased intestinal glucose absorption by 25.42%. No adverse effects were seen in mice administered CPSO at up to 5 ml kg À1. Conclusion: CPSO is antihyperglycemic. The effect can be explained partly by inhibition of intestinal glucose absorption.

Objective: Extraction and evaluation of the antidiabetic activity of extracts from stem and roots of Nerium oleander (Apocynaceae) Linn.Methods: Stem and roots of N. oleander were collected, dried and extracted by using well-established methods for alkaloids, flavonoids, steroids and crude extracts in polar and non-polar solvents. Evaluation of their antidiabetic activity was done with salivary alpha-amylase and starch as a substrate using chromogenic DNSA (2,4-Di nitro Salicylic Acid) method and Starch-iodine method. All experiments were performed in 3 different sets each in triplicates. The data are expressed as mean±SEM (standard error of the mean). Results:The highest inhibition for stem was found in its free flavonoid extract at the concentration of 1.5 mg/ml, with percent inhibition 48.35±1.36 % and an IC50 value of 1.774 g/ml while in case of root, highest inhibition was obtained at 1.5 mg/ml of pet ether extract, with % inhibition 52±0.40 % and IC50 value 1.583 g/ml and at 1.5 mg/ml of methanol extract, with % inhibition 42.12±1.12 % and an IC50 value 1.729 g/ml. 8 (5 of stem and 3 of root) out of 14 tested extracts have shown good inhibitory potential. Extracts of the stem were found to be more potent than root extracts. Conclusion:Though stem extracts were found to be a more potent hypoglycemic agent than root extracts, however, extracts of both parts have good antidiabetic potential and both might be fruitful in managing the postprandial hyperglycemia.
