São Paulo.OBJECTIVES: to evaluate the toxicity of the hydrogel dressing with nano-silver at 22 ppm and 44 ppm in superficial skin lesions using Wistar rats, from the histopathological analysis of part of the liver and kidneys; evaluate the possible toxic effects on kidney, liver and blood cell functions through the blood count. METHODOLOGY: This is a preclinical study. The hydrogel and hydrogel dressings with nano-silver with concentrations of 22 and 44 ppm were produced at the Chemistry and Environment Center (CQMA) of IPEN. The research was carried out at the Vivarium of Faculdade Presidente Antônio Carlos in Porto Nacional (FAPAC ITPAC Porto), Tocantins. Eightyfive young adult male rats were used, with an average age of 60 to 90 days. The animals were randomly classified into three groups. The lesions were treated every three days and once a day. The clinical signs and body weight of the rats were recorded. Blood samples and organs were later collected for biochemical examination.The anatomical specimens, after surgically removed, were submitted to routine blocking and histotechnical procedures. The project was approved by the FAPAC ITPAC Porto Animal Use Ethics Committee. All data were tabulated and analyzed using the GraphPad Prism version 8.0.1 software, Shapiro-Wilk test to verify the normality of the data. For non-parametric data, the Kruskal-Wallis test and comparisons using the Dunn post-test were used. For normal distribution data analysis (ANOVA) and through the Tukey test. RESULTS: For each hematological parameter evaluated at different times, the three groups behaved in a very similar way and demonstrated that they were not statistically different, with few exceptions to what refers to mononuclear blood cells. In the analysis of urea dosage the means of the control group compared to those of the other treatments, it was noted that the means of the HP22 were higher in the time of 7 and 14 days, while the group of the HP44 were higher in 24 hours, 7 days and 14 days. In the creatinine dosage, only the group (HP44) 14 days was shown to be higher. When comparing the means of ALT, in relation to the group (H), it was observed that the means of (HP22) were higher in 24 h and 3 days, while the group (HP44) showed to be higher in 24 h, 7, 14 and 30 days.In AST it was noted that (HP22) was higher in 3, 7 and 14 days while (HP44) was higher in 24 h and 7 days. The study did not show any significant changes regarding the histopathological analyzes of the liver and kidneys. CONCLUSIONS: hydrogel dressings with crosslinked silver nanoparticles and sterilized by gamma rays produced at IPEN / USP, did not induce toxicity to animals and the data will serve as a basis to estimate the potential risk of possible exposure to human health.
