This research describes the preparation of membranes with chitosan (CS) as the polymeric matrix and cellulose nanocrystals (CNC) as reinforcement. The aim was to evaluate their physical, mechanical and biological properties, and to determine their potential for biomedical use. Membranes were prepared via casting CNC suspensions in CS solution, at CNC concentrations of 0.5%, 1.0% and 2.0% (w/w) with pure chitosan as a reference. Analysis of membrane properties was performed using several techniques, such as ATR-FTIR, SEM, swelling test, maximum water absorption, dynamical mechanical analysis and in vivo (Winstar rats) biocompatibility and biodegradability assays for biological evaluation. Experimental results established that CNC reduced swelling rates and increased the maximum water absorption when CNC concentration was higher. Therefore, the presence of CNC in the matrix reduced Young's modulus by approximately 50% in comparison with pure chitosan. All formulations demonstrated biocompatibility and biodegradability values ranged between 4% and 21% in the 30 days after implantation. Based on these results, these membranes may be of use for biomedical applications.

Evaluation of Local Tissue Reaction After the Application of a 3D Printed Novel Holdfast Device for Left Atrial Appendage Exclusion

The left atrial appendage (LAA) is a small, fingerlike extension of the left atrium and its exclusion is used as a treatment strategy to prevent ischemic stroke. Existing holdfast devices may damage the tissue, are unisized and not adjustable. A novel holdfast device for LAA exclusion devoid of these shortcomings was designed and 3D-printed using the Selective Laser Sintering (SLS) technology with polyamide powder and tested it on animal model. We selected the SLS 3D printing technology due to its wid14e availability and low production costs which could provide on-site 3D printing for specific patient. The purpose of this study was to evaluate the biocompatibility of the reported holdfast device and compare the histological results obtained for local tissue reactions to those obtained for an established grafting material. Thirty swine subdivided into two groups were examined. The LAA exclusion device was implanted and was either coated with a polyester vascular implant or not coated at all and the histological response to the device's presence was evaluated which is a standard approach to test the device biocompatibility. In all cases, complete occlusion was seen without any pathological findings during the incubation time. In both groups, the surface of the atrium under a holdfast device was smooth and shiny and had no clots. The foreign body reaction of the LAA holdfast device made of polyamide powder was insignificantly lower compared to the polyester graft. Thus, it fulfils the parameters of biocompatibility at the highest degree, and makes it suitable material for the manufacturing of LAA holdfast devices.

Since the beginning of the human race, there was those who sought to assist the body in a natural attempt to restore yours injured parts. One of the main current challenges is the replacement of body tissues, including areas of skin lesion. A biomaterial can be used to improve, enhance or replace, partially or fully tissues or organs. The membrane of bacterial cellulose (BC) has moldability, good mechanical properties, selective permeability, allowing the passage of water vapor but preventing the passage of microorganisms. The collagen (COL) has been widely used as material in the manufacture of biomaterials. In this study was obtained hydrogel and membrane BC-COL, characterized in different ways. Were realized in vivo studies, macroscopic and histological analyzes from dressings of BC-COL, comparing with controls (clot and collagenase ointment), after applying in wounds on the backs of rats. The animals were sacrificed after 3, 7, 15 and 30 days, and the scars were processed according to histological routine to HE staining. The characterizations performed in this study (scanning electron microscopy (SEM), thermogravimetric analysis (TGA), infrared spectroscopy with Fourier transform (FT-IR) and X-ray diffraction (XRD)) confirmed the incorporation of the COL to matrices BC. The macroscopic evaluation only demonstrated statistically significant difference of tissue repair between treatments at seven days postoperative, and the hydrogel showed a trend for a faster repair. The results of the histological evaluation showed statistically significant difference in inflammatory tissue reaction between treatments in all periods studied. In quality evaluation, quantity and orientation of collagen fibers, only three days period didn't show statistically significant difference between treatments. We conclude from these results that the two dressings are biocompatible.
