Estrus synchronization schemes and application efficacies in anestrus lanzhou fat-tailed ewes

Abstract: This study aims to investigate the estrus efficacy of FIS (fluorogestone intravaginal sponges)/eCG (equine chorionic gonadotropin) and cloprostenol treatments to establish the optimum protocols for estrus synchronization in ewes. Forty-two ewes were assigned into groups A (FIS/eCG group) and B (CLO (cloprostenol) group). Each group was randomly divided into three subgroups (n = 7) according to the dose of eCG (ECG1, ECG2 and ECG3) or cloprostenol (CLO1, CLO2 and CLO3). The estrus rates, lambing rates and avera… Show more

“…A total RNA of 30 COCs was extracted from cumulus cells after IVM (during which cells were exposed to IVM medium containing various FSH and LH concentrations) using TRIzol reagent (Invitrogen, Beijing, China) according to the manufacturer's instructions, before being reverse transcribed with PrimeScript TM RT reagent kit (TaKaRa Co., Dalian, China) according to the manufacturer's instructions (Wei et al, 2013;Wei et al, 2016). The amount of 100 μg RNA was used for the experiment.…”

“…However, currently little information has been reported on LH effects on the IVM of ewe oocytes (Xiao et al, 2014). To date, it is unclear whether supplementation of FSH or LH into IVM medium affects maturation and apoptosis of oocytes in Lanzhou fat-tailed sheep (de Frutos et al, 2013;Wei et al, 2016), or which dose of FSH and LH in IVM medium is the optimum of IVM of oocytes in this breed of sheep.…”

“…Induction of estrus with an intravaginal progesteronereleasing device (IPRD) and GnRH can enhance pregnancy rates in adult ewes, and ewe lambs out of the breeding season [3] . Exogenous gonadotrophins, such as pregnant mare serum gonadotrophin (PMSG), are used to stimulate follicular growth, which increase the estrus response and ovulation rate [4,5] , and also lead to a tighter synchrony of ovulation in out-of-season estrus induction and cyclic ewes [6,7] .…”

“…Equine chorionic gonadotropin [eCG, previously known as pregnant mare serum gonadotropin (PMSG)] demonstrates an efficacy similar to those of FSH and LH, and has a high affinity with the receptors (FSHR and LHR, respectively) of both of these hormones in the ovaries. eCG administration can improve reproductive performance and increase ovulation and pregnancy rates in noncyclic cows (Uslu et al, 2012;Hashim et al, 2013) and ewes (Wei et al, 2016a). However, the effect of supplementing IVM medium with eCG on IVM, apoptosis, and the expression and mRNA levels of reproductive hormone receptors in Lanzhou fat-tailed sheep remains unknown (Karami-Shabankareh et al, 2015).…”
