The present study compared the effects of follicle stimulating hormone (FSH) and luteinizing hormone (LH) on in vitro maturation (IVM), apoptosis, and secretion function in sheep oocytes, as well as gene expressions of the receptors (FSHR, LHR, and GnRHR) in cumulus-oocyte complexes (COCs). The COCs were recovered from sheep ovaries and pooled in groups. The COCs were cultured for 24 hours in IVM medium supplemented with various concentrations of LH (5-30 μg/mL) and FSH (5-30 IU/mL). They were allocated to LH-1 (5 µg/mL), LH-2 (10 µg/mL), LH-3 (20 µg/mL), and LH-4 (30 µg/mL) groups, and FSH-1 (5 IU/mL), FSH-2 (10 IU/mL), FSH-3 (20 IU/mL), and FSH-4 (30IU/mL) groups. The apoptosis of COCs was assessed by terminal deoxynucleotidyl transferase dUTP nick end labelling (TUNEL). The maturation rates of oocytes improved gradually as LH and FSH concentration increased from 0 to 10 μg/mL(IU/mL), reaching a peak value of 44.1% of LH-2 and 48.5% of FSH-2 group. Oocyte apoptosis rates of LH-2 and FSH-2 groups were the lowest among LH-and FSH-treated groups, respectively. The germinal vesicle breakdown (GVBD) rate of the FSH-2 group was higher than that of the control group (CG) and FSH-4 groups. The GVBD rate of LH-2 group also increased in comparison with the CG group. FSH concentration of the FSH-2 group was greater than that of CG. Expression levels of FSHR, LHR, and GnRHR mRNAs of FSH-2, LH-3, and LH-3 group, respectively, were higher than CG. Levels of FSHR proteins in FSH-2 and FSH-3 groups were greater than CG. Levels of GnRHR proteins were increased with a maximum increment of FSH-4. The FSH and LH supplemented into the IVM medium could promote the maturation rate, reduce the apoptosis rate of sheep oocytes, and increase FSH concentrations in IVM medium fluid. Additionally, FSH and LH enhanced expression levels of FSHR, LHR, and GnRHR mRNAs of sheep COCs.

Induction of estrus is an effective management tool for increasing the pregnancy rate in ewes, and there is a long estrus interval after lambing in spring in Small-tail Han ewe. In this study, norgestrel releasing intra-vaginal devices (PRID) and PRID + pregnant mare serum gonadotrophin (PMSG) were used for estrus induction in Small-tail Han ewe, and expression of follicle-stimulating hormone receptor (FSHR), luteinizing hormone receptor (LHR), estrogen receptor (ER) and progesterone receptor (PGR) mRNA in the ovarial stroma, oviduct ampulla and endometrium was analyzed by quantitative real-time PCR. The results showed that the estrus-induction with PRID and PRID + PMSG almost had the same effects on increasing the percentage of animals in estrus and conception rate of the ewes, and the estrus-induction had no significant effect on the expression of LHR, ER and PGR mRNA in the oviduct ampulla, ovarial stroma and endometrium, and FSHR in the ovarial stroma and endometrium. Furthermore, the estrus-induction could significantly decrease the expression of FSHR mRNA in the ovine oviduct ampulla (P<0.05). Therefore, it was suggested that down-regulation of FSHR in the oviduct ampulla after estrus induction may be helpful for fertilization and early embryo development.

We assessed the effects of equine chorionic gonadotropin (eCG) on oocyte in vitro maturation (IVM), apoptosis, and follicle-stimulating hormone receptor (FSHR), luteinizing hormone receptor (LHR), and gonadotropin-releasing hormone receptor (GnRHR) expression and mRNA levels. Cumulus-oocyte complexes (COCs) were recovered from sheep ovaries and pooled in groups, before being cultured in IVM media containing varying eCG concentrations. Maturation and apoptosis rates were then calculated. Expression of FSHR, LHR, and GnRHR mRNA in oocytes was measured using quantitative reverse transcription polymerase chain reaction. Protein levels were ascertained by western blotting. Matured oocytes displayed and released an intact first polar body. Sheep oocyte maturation rates gradually increased as eCG concentration was raised from 0 to 20 µg/mL. Apoptosis rates of eCG-treated oocytes were lower than those of the control group, and were lowest using 20 µg/mL eCG. FSHR, LHR, and GnRHR mRNA expression increased (P < 0.01, P < 0.05, and P < 0.05, respectively, compared to 0 µg/mL eCG) with eCG concentration, being highest following exposure to 20 µg/mL. FSHR and GnRHR protein levels were significantly higher in oocytes administered 20 µg/mL eCG compared with those matured in the absence of eCG. eCG dose positively correlated with FSHR, LHR, and GnRHR mRNA and protein expression. In conclusion, eCG enhances maturation and decreases apoptosis of oocytes undergoing IVM, and heightens FSHR, LHR, and GnRHR expression. Such increased expression may facilitate oocyte IVM. These findings contribute to our understanding of the mechanisms of underlying hormonal control of sheep oocyte IVM, advancing ovine reproductive methods.
