Estrogen‐TGFβ cross‐talk in bone and other cell types: Role of TIEG, Runx2, and other transcription factors

Abstract: It is well established that E 2 and TGFβ have major biological effects in multiple tissues, including bone. The signaling pathways through which these two factors elicit their effects are well documented. However, the interaction between these two pathways and the potential consequences of cross-talk between E 2 and TGFβ continue to be elucidated. In this prospectus, we present known and potential roles of TIEG, Runx2, and other transcription factors as important mediators of signaling between these two pathwa… Show more

“…Runx2, a transcription factor, was reported to participate in the processes of osteoblast growth and differentiation. (46) This study showed that exposure of osteoblasts to oxidative stress inhibited Runx2 mRNA expression, but pretreatment with NO decreased such inhibition. In heavy metal-induced osteotoxicity, cadmium inhibited Runx2 mRNA expression and triggered apoptotic death of cells of the Saos-2 human osteoblast-like cell line.…”

“…Runt-related transcription factor 2 (Runx2), an osteogenic master transcription factor, was reported to regulate osteoblast differentiation and maturation. 12,13 Previous studies showed that activation of Runx2 induces ALP, OCN, and OPN messenger (m)RNA expression and concurrently promotes osteoblast differentiation. 14,15 Thus, Runx2-involved regulation of osteoblast-associated gene expression makes a proportionally major contribution to regulating osteoblast proliferation and maturation.…”

“…Runx2 functions as an osteogenic master transcription factor and regulates osteoblast differentiation and maturation. 12,13 The p300/CBP-associated factor is reported to activate Runx2 by directly binding to and acetylating this transcription factor.…”

“…Runt-related transcription factor 2 (Runx2) is implicated as an essential transcription factor for osteoblast differentiation and mineralization. 25, 26 Wohl et al reported that Runx2 expression is associated with osteogenesis and bone repair. 27 Moreover, alkaline phosphatase (ALP) and osteocalcin (OCN) are two typical osteoblast biomarkers that participate in controlling osteoblast function and ECM mineralization in osteogenesis and bone remodeling.…”
