Estrogen influences satellite cell activation and proliferation following downhill running in rats

Abstract: To investigate the influence of estrogen on postexercise muscle repair processes, we examined the effects of estrogen supplementation (0.25-mg pellet) on numbers of myofibers positive for markers of total, activated, and proliferating satellite cells in rat skeletal muscles 72 h following downhill running. Ovariectomized female rats (n = 44) were divided into four groups (n = 11 per group): sham (no estrogen) controls (SC); sham, exercised (SE); estrogen-supplemented controls (EC); and estrogen-supplemented, e… Show more

“…There were no differences (P Ͼ 0.05) in body weight between any of the groups at Pre, whereas, at Post, body weight was lower (P Ͻ 0.05) in E compared with both S and P. There were no differences (P Ͼ 0.05) in body weight between S and P at Post. The lower body weights observed for E relative to S are in accordance with previous studies using ovariectomized rats (6,26,42). The plasma concentrations of estradiol and progesterone of all treatment groups are also shown in Table 1.…”

“…Animals were returned to their cages and allowed to recover for 8 days before beginning the exercise protocol. We have demonstrated previously that 1 wk of estrogen exposure in this model is sufficient to allow estrogen effects on skeletal muscle to manifest (6,43). This recovery time allowed the pellet-implanted groups to receive prolonged exposure to the ovarian sex hormone.…”

“…Recent evidence suggests that oestrogen receptors (ERα and ERβ) within the muscle can stimulate the proliferation of satellite cells via the PI3K/Akt pathway for muscle growth and repair 33,40 . EE increases the proliferation of satellite cells in rat muscle tissue 41 , indicating that the synthetic component of hormonal contraceptives may have a direct anabolic e ect on muscle, possibly due to a local activation of IGF-1 pathway through an autocrine or paracrine manner 42 .…”
