Estrogen-dependent effects of 5-hydroxytryptophan on cortical spreading depression in rat: Modelling the serotonin-ovarian hormone interaction in migraine aura

Abstract: Background: Cortical spreading depression (CSD) is the likely culprit of the migraine aura. Migraine is sexually dimorphic and thought to be a "low 5-HT" condition. We sought to decipher the interrelation between serotonin, ovarian hormones and cortical excitability in a model of migraine aura.

“…Administration of the serotonin precursor 5-hydroxytryptophan in animal models has also been shown to decrease the frequency of KCl-induced SD in female rats, suggesting a possible role for serotonergic agents as therapeutic tools in migraine with aura in females ( 109 ). Blocking the SD mechanism, therefore, seems to be a rational approach to explore in molecular research aimed at improving the prevention and treatment of migraine with aura.…”

“…These findings may explain why migraine aura can emerge with pregnancy as estrogen levels rise and why MRM is rarely associated with MA. 39 This study built upon work which had demonstrated that enzymes involved in the synthesis of serotonin are expressed in mouse trigeminal ganglion, and the level of these enzymes is dependent on the gender and estrus cycle stage, suggesting that ovarian steroids might play a role in the regulation of serotonin synthesis. 40…”

“…In high oestrogen states, such as pregnancy and during use of the combined oral contraceptive pill or high-dose hormone replacement therapy, aura can be triggered; conversely, a decline in oestrogen concentration is associated with exacerbations of migraine without aura, probably by reducing the migraine attack threshold or by directly triggering attacks 9. Animal studies support the association between high oestrogen states and aura in that oestrogen increases the susceptibility to cortical spreading depression, the pathophysiological correlate of migraine aura 10. This phenomenon is thought to be glutaminergic 11…”
