Estradiol, Progesterone and Testosterone Exposures Affect the Atrial Natriuretic Peptide Gene Expression in vivo in Rats

Abstract: To clarify the effects of sex hormones on the expression of atrial natriuretic peptide (ANP), ovariectomized and intact female rats were subcutaneously injected with estradiol, progesterone, a mixture of them or olive oil solvent; castrated and untouched male rats were subcutaneously injected with estradiol, testosterone or olive oil, once a day for 7 days. The relative rANP-mRNA contents of rat atrial were measured by molecular hybridization. rANP-cDNA was labeled with 32 P as a probe. The results revealed tha… Show more

“…16,17 Furthermore, it has been shown previously that E 2 increases the expression of ANP. 14,28 In concordance with these results, E 2 led to an increase in ANP expression in the ventricles of animals with pressure overload compared with placebo. These findings suggest that the antihypertrophic effect of E 2 may be mediated by the increased expression of ANP.…”

“…One murine-based study demonstrated that treatment with estradiol and progesterone stimulated NP gene expression in females (30 ), while another study demonstrated that testosterone suppressed volumestimulated NP release from isolated perfused rat atria (31 ). Clinically, a study of postmenopausal women demonstrated a significant increase in mean BNP concentrations after transdermal estradiol administration (32 ), while observational studies have identified inverse correlations between circulating androgens and NP concentrations (Fig.…”

“…The activation of these receptors leads to a reduction in preload by the elimination of water and sodium and by shifting plasma volume from intravascular to extravascular spaces 20 . The natriuretic and diuretic actions of NPR-A result from enhanced glomerular filtration and/or decreased tubular reabsorption of sodium and water, suppression of renin, aldosterone, and vasopressin secretion as well as antagonism of most of the peripheral and central effects of angiotensin II 19,21 . If the chronic training promotes a parallel increase of NPRs and NPs, then this also explains the concomitant increased production of NO and NPs 22 , observed by Gutkowska et al 14 .…”
