Estradiol Dose-Dependent Regulation of Membrane Estrogen Receptor-α, Metabotropic Glutamate Receptor-1a, and Their Complexes in the Arcuate Nucleus of the Hypothalamus in Female Rats

Abstract: Sexual receptivity in the female rat is dependent on dose and duration of estradiol exposure. A 2 μg dose of estradiol benzoate (EB) primes reproductive behavior circuits without facilitating lordosis. However, 50 μg EB facilitates lordosis after 48 hours. Both EB doses activate membrane estrogen receptor-α (mERα) that complexes with and signals through metabotropic glutamate receptor-1a (mGluR1a). This mERα-mGluR1a signaling activates a multisynaptic lordosis-inhibiting circuit in the arcuate nucleus (ARH) th… Show more

“…ARH, VMH, and posterodorsal medial amygdala [41]). FG retrograde labeling was observed through the rostrocaudal extent of the ARH as reported previously [4], primarily in the dorsal to ventral extent of the medial portion of the ARH as well as the mediolateral extent of the ventral ARH.…”

“…Protein analysis and ARH blocked dissection were performed as described in Mahavongtrakul et al, [41]. Briefly, brains were quickly removed from the cranium, chilled in 4°C PBS and block dissections of the ARH 2 mm thick were collected into 1.5 ml Eppendorf tubes.…”

“…Interestingly, various doses of estradiol regulate mERα signaling by regulating its levels on the membrane [107]. Rats given a low dose of EB continue to have mERα-mGluR1a in the ARH.…”

“…Treating EB-primed (2 μg) nonreceptive rats with ERα/β antagonists (ICI 182,780 or tamoxifen) facilitates lordosis (Garcia et al, 2010). This could be through down regulation of ARH membrane ERα-mGluR1a complexes to reduce β-endorphin release and deactivate MPN MOR (Mahavongtrakul et al, 2013). However, these ER antagonists also activate G protein-coupled estrogen receptor-1 (GPER; aka GPR30 reviewed in Micevych and Sinchak, 2013; Prossnitz et al, 2008; Sinchak and Wagner, 2012).…”
