Japanese pear (Pyrus pyrifolia Nakai) is an important Rosaceous fruit tree in Japan. This species exhibits gametophytic self-incompatibility (GSI), which is controlled by a single S-locus with multiple S-haplotypes. Although GSI is a genetic mechanism to prevent inbreeding and promote outcrossing to maintain genetic diversity, it can be problematic in fruit trees because it causes unstable fruit set. Therefore, orchardists interplant Japanese pear with other lines as pollinizers or conduct artificial pollination to ensure fruit set. To achieve stable fruit production and to reduce or eliminate the need for artificial pollination, research on the GSI of Japanese pear has been conducted by pollination experiments and by characterization of selfcompatible (SC) mutants. Additionally, breeding programs have progressed to produce SC cultivars with high fruit quality. Recently, molecular analyses of GSI and SC mutants in Japanese pear have provided new information that is relevant to the stable fruit production and efficient breeding of Japanese pear. This review focuses on studies of the GSI of Japanese pear, and especially on the recent development of DNA markers for S-genotyping and marker-assisted selection of SC trees. In addition, the candidate genes controlling pollen S specificity and a model of the molecular mechanism of GSI in Japanese pear are described.

Most rosaceous fruit tree species exhibit gametophytic self-incompatibility (GSI). During the last decade, much progress has been made in identifying and characterizing the S-locus genes that control the specificity of the selfincompatible (SI) reaction in the Rosaceae. The pistil and pollen determinants of S-specificity in the Rosaceae are a ribonuclease and an F-box protein, respectively. From allele-specific sequence polymorphism of S-specificity genes, molecular S-genotyping systems have been established in various rosaceous fruit tree species. Molecular characterization of S-determinants in self-compatible (SC) cultivars has revealed defects in the S-determinants that cause SC and enabled the development of molecular markers for SC. Marker-assisted selection of SC offspring is now being successfully incorporated into breeding programs. In this paper, we summarize the current knowledge of the molecular basis of self-(in)compatibility, S-genotyping methods, and molecular markers for SC in rosaceous fruit tree species.

S-RNase alleles and 18 simple sequence repeats (SSR) markers were used to analyze 55 Japanese pear cultivars, including 47 hybrid cultivars, 6 bud sport mutants, and 2 derived from open pollinations. Fifteen cultivars used in the breeding of some of the hybrid cultivars were also examined. Each cultivar was genotyped at the S-locus and SSR loci. Parent-offspring relationships were analyzed by comparing genotypes among the cultivars. Of the 47 crossbreeding cultivars and selections, parent-offspring relationships of 37 were re-confirmed because the hybrids inherited S-RNase and SSR alleles from both parents without any discrepancies.
