Estimation of phenolic conjugation by colonic mucosa.

Abstract: SUMMARY Conjugation of phenol by the colonic mucosa was assessed in vivo using dialysis tubing containing 15 ml of 1 mmol/l acetaminophen (paracetamol) and 10 mmol/l butyrate. These were allowed to equilibrate in the rectum for one hour. The glucuronidated and sulphated conjugates of acetaminophen were measured by high pressure liquid chromatography and bicarbonate concentrations by gas analysis. In 21 subjects without colonic disease sulphate conjugation was observed in all cases, with a mean (SE) of 3-86 (0-… Show more

“…PAG and 4CS are formed from protein putrefaction of phenylalanine and tyrosine by the gut microbiota. Phenylalanine is converted to phenylacetate in the colon and subsequently conjugated with glutamine in the liver and the gut mucosa 53 , whereas 4CS is a product of microbial tyrosine breakdown via hydroxyphenylacetate to 4-cresyl , followed by conjugation with sulfate. 54 Age-related variations were also observed in the bacterial fermentation product, lactic acid, being negatively associated with aging in the American sample.…”

“…A likely explanation for this is that the formula feeds currently available, including the products used in the present study, contain similar amounts of tyrosine (the substrate from which gut bacteria generate phenol and p-cresol) to levels found in breast milk. Phenols are usually detoxified by either sulfate or glucuronide conjugation in the colonic mucosa and then excreted in the urine or remain unabsorbed and excreted in the faeces (Ramakrishna et al 1989).…”

“…3). As regards the site of this competitive sulfonation, the colonic mucosa is known to have significant sulfonation capacity (25,31) and could potentially convert colonically produced p-cresol to PCS. However, with acetaminophen being rapidly absorbed from the small intestine and with the liver being regarded as the principal site of its metabolism (17), we envisage that some colonically produced p-cresol may escape further colonic modification and be sulfonated in the liver rather than in the gastrointestinal tract.…”
