Tea plants (Camellia sinensis L.) are phenol-accumulating crops that are widely used for public health. The healing effect of tea leaf products is due to the biosynthesis of such phenolic compounds (PCs) as flavans, which have P-vitamin capillary-strengthening activity. Due to their limited habitat and the value of their specialized metabolites of a phenolic nature, a promising approach is to establish in vitro cultures from them that retain the ability to form PCs, which is characteristic of ex vivo tea plants. The aim of this study was to investigate the effect of exogenic H2O2 (0.01 mM; 0.1 mM; 1 mM) on the growth, morphology, degree of stress response, and accumulation of various phenolic compounds in tea plant callus cultures of different ages (24 or 36 days) grown under different cultivation conditions (darkness or light). According to the results obtained, the H2O2 effect on tea callus cultures of different ages did not cause changes in their morphophysiological characteristics, both after 2 h of exposure (rapid response of callus culture, RRCC) and after 48 h (delayed response of callus culture, DRCC). The determination of the malondialdehyde (MDA) content, which serves as an indicator of changes in the level of lipid peroxidation (LPO) and the presence of stress responses in plant cells, indicated either its maintenance at the control level, a decrease, or an increase. All these effects depended on the growth conditions of the tea callus cultures (darkness or light), their age, the duration of exposure (rapid or delayed response), and the H2O2 concentration. Similar trends were noted for the total content of PCs as well as the amount of flavans, proanthocyanidins (soluble and insoluble forms), and lignin. The plant cell responses reflected changes in its adaptation programs, when specialized metabolites act as a target for the action of H2O2, thereby contributing to an increase in their resistance.

Low-temperature storage is convenient for postharvest preservation of peach fruit, but peach fruit is sensitive to cold damage, which lowers its quality. Nitric oxide (NO) has the potential to improve the bitter resistance of peach fruit. In this work, peach fruit was treated with 15 μmol L −1 NO and 5 μmol L −1 c-PTIO [2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxo-3-oxide], to study changes in mitochondrial fatty acids and expression of the C-repeat binding factor (CBF). The results showed that 15 μmol L −1 exogenous NO significantly maintained fruit quality, reduced peroxidation of mitochondrial fatty acids, increased the activities of the antioxidants superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbic acid peroxidase (APX), and reduced the content of hydrogen peroxide (H 2 O 2 ). Meanwhile, NO treatment suppressed the increase in browning index and ion leakage rate, increased the activity of S-nitrosoglutathione reductase (GSNOR), the contents of S-nitrosothiols (SNOs), and the ratios of mitochondrial NAD + /NADH and NADP + /NADPH, increased the expression levels of PpCBF1/5/6. However, the expression levels of PpCBF2/3/4 were not significantly regulated by exogenous NO. Peaches treated with c-PTIO showed opposite effects to those treated with exogenous NO. These results suggest that exogenous NO can improve antioxidant capacity, preserve mitochondrial fatty acids, and upregulate the expression of PpCBF1/5/6 to alleviate cold tolerance and maintain the peach quality during storage.

Background: In India, nicotine addiction causes a huge socio-economic burden, mortality, and morbidity. Chronic intake develops a variety of symptoms and life-long medical complications. The study aims to examine the nicotine-induced effects on cellular oxidative stress and psychological stress and its correlations with homocysteine (Hcy) concentration in nicotine-addicted respondents. Methods & Results: Nicotine-induced psychological stress was assessed through perceived stress scales (PSS) and coping self-efficacy (CSE) scales and oxidative stress was estimated through the markers- Malondialdehyde (MDA), superoxide dismutase (SOD), and Catalase. We found significantly higher (p=0.0001) perceived stress (PS) with diminished CSE in addicted respondents whereas low PS with significantly increased (p=0.0001) CSE were observed in non-addicted. In erythrocytes (RBCs), MDA content was found significantly increased (p=0.0006) in the nicotine-addicted group that may cause oxidative damage. SOD activity was significantly decreased (p=0.0001) in the addicted group. Similarly, plasma Catalase activity was also decreased (p=0.02). Nicotine intake influences Hcy concentration, by 55%, 27.5%, and 7.5% in addicted respondents categorized as moderate, intermediate, and severe Hcy categories. Conclusions: The minimal short-term nicotine consumption has the antioxidant role that promotes positive stimulus on memory and triggers executive functions but the same gets impaired and mitigated on chronic exposure. Chronic nicotine consumption induces oxidative stress and perceived psychological stress as well as influences the Homocysteine concentration in the addicted individuals. Higher oxidative stress impairs cellular defensive mechanisms and causes the risk for severe damage to cells and tissues.
