Estimation of ethylene production and 1-aminocyclopropane-1-carboxylic acid in plants by means of gas chromatography

Abstract: The paper deals with problems associated with preparation and collection of samples when estimating the production of ethylene and content of ACC (1-aminocyclopropane-1-carboxylic acid) in plants by means of gas chromatography. A proper method of sampling can significantly influence not only the reliability of obtained data but also their interpretation. Attention was paid to cultivation of plant material, sampling vessels, conditions of ethylene production, sampling procedure, and storage of gaseous samples. … Show more

“…After 3 days of germination, 1 ml of gas was taken off from the cultivation vessel through a plastic tuberculin syringe placed in the vessel cap. Ethylene content was analysed in a gas chromatograph (GC800 Series with 50 m capillary column Al 2 O 3 "S" 15 mm, ID 0.53 mm, Fissons Instrument, Italy); when estimating gaseous hydrocarbons, spray, column and detector temperatures were 230ºC, 40ºC and 200ºC, respectively [24].…”

“…samples stored in the atmosphere without an increased level of ethylene) took place simultaneously in another micro malt house with identical internal conditions. The content of oxygen was measured with an oxygen electrode in the digital oxymetre GMH 3691, while contents of carbon dioxide and ethylene were estimated by means of gas chromatography [15,16,9]. je způsobeno časovým rozdílem odběru vzorků od ukončení větrání mikrosladovny.…”

“…Sladování kontrolní varianty (bez zvýšené hladiny ethylenu) probíhalo souběžně za stejných podmínek v druhé mikrosladovně. Obsah kyslíku byl měřen kyslíko-vou elektrodou digitálním oxymetrem GMH 3691, obsah CO 2 a ethylenu byl stanoven pomocí plynové chromatografie [15,16,9].…”
