Estimation of collagen content of liver specimens. Variation among animals and among hepatic lobes in cirrhotic rats.

Abstract: We undertook a study to evaluate the correlation between morphometric evaluation and cobonimetnic determination of hepatic collagen content, and to analyze the variation among animals as well as among lobes of the same liver in hepatic collagen content after CC14-induced micronodulan cirrho-sis_ The results revealed a significant correlation (r = 0.9458; p < 0.001) between the morphometric and colonimetric methods of collagen evaluation of liver specimens; both methods also significantly distinguished data obt… Show more

“…By this procedure collagen is stained red. 40 For hybridization, the sections were deparaffinized in xylene, rehydrated through a graded series of ethanol solutions, rinsed in distilled water (5 minutes), and incubated in 2 ϫ sodium saline citrate at 70°C for 30 minutes. The sections were then rinsed in distilled water and treated with pronase (0.125 mg/mL in 50 mmol/L Tris-HCl, 5 mmol/L ethylenediaminetetraacetic acid, pH 7.5) for 10 minutes.…”

“…The presence of total collagen in the granulation tissue was analyzed using Sirius red and fast green staining as previously described (Gascon-Barre et al, 1989;LopezDe Leon and Rojkind, 1985;Peterson, 1993). Briefly, tissue sections were incubated in 0.04% fast green in saturated picric acid for 15 minutes at room temperature.…”

“…One week after the last injection, mice were sacrificed and serum transaminase activities determined (e and f). The degree of fibrosis was determined by measurement of the amount of collagen per milligram of total liver proteins as previously described (24). The differences between the two groups after Con-A injections are statistically significant (P < 0.05) for AST and ALT, but not for collagen.…”

“…To test this possibility, we also injected mice multiple times with a low dose of Con-A. Liver fibrosis was assessed by both collagen assay and histochemistry (24,25). Interestingly, as shown in Figure 2, TRAIL defi-…”
