Estimating the intra- and inter-individual imprecision of manual pipetting

Abstract: The results of this study show that manual pipetting is plagued by a considerable intra- and inter-individual imprecision, which is inversely correlated with the amount of fluid dispensed.

“…injury exposure, unreliable cold chain, prolonged transportation time), presence of interfering substances (e.g. release of cellular components that may interfere with the assay due to whole blood freezing, use of inappropriate additives) [13][14][15], as well as by a number of procedural issues occurring during sample preparation, thus including pipetting errors during manual sample preparation or aliquoting, cross-contamination and sample mismatch, among others [16]. The leading specific problems that may plague the quality of RT-PCR assays include sample contamination (even trace amounts of external DNA may jeopardize test results) and testing carried out in patients receiving antiretroviral therapy, which may then generate falsenegative results [15].…”

“…Although microplate ELISAs are very sensitive to Ddimer and have long been considered the reference method [1][2][3], they are manual, require technical skills, are time-consuming (i.e. around 2-4 h), and are plagued by a high degree of analytical imprecision [2,83].…”

“…We read with interest articles written by Kitchen The innovative technology of the cobas t 711 analyzer provides some advances over existing coagulation instrumentation, such as availability of lyophilized reagents in barcoded cassettes, which can be automatically resuspended by the analyzer onboard, and may hence be seen a step forward for reducing the inherent imprecision of manual resuspension. 3 This may be especially beneficial in stat clinical laboratories, for shortening the turnaround time (TAT) and improving laboratory efficiency by decreasing manual activities. 4 The availability of rapid and accurate D-dimer (DD) assays is essential for timely management of patients with many thrombotic disorders, especially for ruling out venous thromboembolism (VTE) and disseminated intravascular coagulation (DIC).…”
