Establishment of temporomandibular joint puncture technique in rats usingin vivomicro-computed tomography (R_mCT®)

Abstract: The aim of the report was to establish puncture techniques for the temporomandibular joint (TMJ) cavity in rats. The experimental sample comprised 30 male Sprague-Dawley rats. Under general anaesthesia the superior joint cavity of the rat was punctured either laterally (lateral puncture technique (LPT), n 5 11), anteriorly (anterosuperior puncture technique (ASPT), n 5 13) or anteroinferior puncture technique (AIPT), n 5 6) using a 27-gauge needle. After the tip of the needle was confirmed by micro-CT (R-mCT H… Show more

“…A 27-gauge needle was advanced into the TMJ space along the superomedial border of the zygomatic arch until it reached the condyle. This method of administration to the TMJ has been shown to be very accurate according to arthrographic CT of the joint, using contrast media [ 41 ].…”

“…Also, the radiation dose of typical micro-CT scans used for mouse lungs is below 10 to 30 mGy [35], [36], [37], [38], over 50 times less than the values capable of inducing significant lung damage [25]. Therefore, it can thus be repeated in individual animals, i.e., micro-CT enables longitudinal studies.…”

“…The MIA was dissolved in 50 μl of contrast media (Optiray 320, FujiPharma, Fujiyama, Japan) and injected with the lateral puncture technique into the upper compartment of the TMJ using a 27‐gauge 0.5‐inch needle while the rats were under anaesthesia by isoflurane inhalation without surgical assistance. To ensure that MIA was introduced correctly in all rats, we used contrast media instead of saline to confirm that the joint cavity was filled into the upper compartment of the TMJ, using in vivo microcomputed tomography (micro‐CT) postinjection (Kameoka et al., ) (Figure ).…”
