Establishment of method for dual simultaneous detection of PEDV and TGEV by combination of magnetic micro-particles and nanoparticles

Abstract: a b s t r a c tTransmissible gastroenteritis virus (TGEV) and porcine epidemic diarrhea virus (PEDV) are the main pathogens causing viral diarrhea in pig, mixed infections of these two viruses are very common in intensive pig rearing. However, there is a lack of a method to simultaneously detect and distinguish PEDV and TGEV in preclinical levels. In this study, we aimed to establish a dual ultrasensitive nanoparticle DNA probe-based PCR assay (dual UNDP-PCR) based on functionalized magnetic bead enrichment an… Show more

“…In addition, the detection rate of TGEV has been at a low level without a trend of large-scale epidemics, while the detection rate of PoRVA and PDCoV have had a rising tendency in recent years. The detection results of co-infection revealed that PEDV and PoRVA co-infection is currently the most important form of co-infection rather than the previous PEDV and TGEV co-infection [ 35 , 36 ], indicating that the monitoring and prevention of PoRVA should be strengthened. The seasonality for the prevalence of PEDV has been gradually decreasing [ 37 , 38 ], and many provinces showed high incidence throughout the year, suggesting that the prevention and control of the epidemic should be strengthened in the summer and autumn when PEDV was low in the past.…”

“…In addition, owing to the indistinguishable clinical symptoms from the pathological and epidemiological changes caused by porcine enteric coronaviruses, such as the closely related coronaviruses (Luo et al 2020 ; Malbec et al 2020 ; Saif et al 2019 ), transmissible gastroenteritis virus (TGEV), and porcine deltacoronavirus (PDCoV), rapid and accurate diagnostic tools are very important for the prevention and control of the spread of PEDV in pigs.…”

“…TGEV is a porcine α-CoV that is often accompanied by mixed infection with PEDV, which seriously concerns the pig industry, and there is no effective therapy yet [ 40 , 41 ]. In order to explore whether hypericin could be a good therapy candidate for both viruses, we analyzed the available crystal structure of TGEV 3CLpro (PDB: 2AMP) and aligned it with PEDV.…”

“…We found these 3CLpro sequences were highly conserved and shared more than 60% identity with PEDV 3CLpro (Table S1), especially the residues in solution 1 (Met6, Ala122, Ile151, Asn152, and Gln295) and solution 2 (Asn141, Leu164, Glu165, Asp186, Gln187, and Gln191) (Figures 3 and 5A), suggesting that hypericin might also be able to inhibit other α-CoV 3CLpro.
