Establishment of In Vitro Regeneration Protocol for Sabah’s Jewel Orchid, Macodes limii J.J. Wood & A.L. Lamb

Abstract: Habitat disturbance and excessive collection of wild orchids from their natural habitat have threatened many orchids species at risk of extinction. In this study, the in vitro regeneration protocol for Macodes limii, a jewel orchid endemic to Sabah was established. The effects of explant source and plant growth regulators (PGRs) including naphthaleneacetic acid, picloram, 2,4-dichlorophenoxyacetic acid, 6-benzylaminopurine, kinetin, and thidiazuron on the in vitro regeneration capacity of M. limii plantlets we… Show more

“…protocorm-like bodies (PLBs)) has been routinely used for mass commercial production and regeneration of endangered orchids [ 197 ]. Propagation protocols have been established for many species with commercial values and conservation purposes [ 198 , 199 ]. In spite of the advantages, somaclonal variations induced by prolonged clonal propagation and cryptic genetic effects under microenvironment become the major limitations for maintaining certain desired traits [ 200 ].…”

“…Even though a vast majority of orchids are frequently multiplied using tissue culture, Phalaenopsis orchids are one of the few economically significant genera that is reticent and causes difficulties for rapid clonal propagation (Singh et al, 2007;David et al, 2022). In addition to meristem culture, flower stalks, internodal segments of flower stalks, flower stalks with axillary bud, root tip (Park et al, 2002), and leaf segments (Park et al, 1996), numerous tissue culture procedures have been established for Phalaenopsis orchids.…”

“…More recently, a study from Malaysia reported the application of this marker in confirming the genetic stability of the in vitro regenerated plants to the mother plant, Macodes limii J.J. Wood & A.L. Lamb., a jewel orchid endemic to Sabah, Malaysia (David et al 2022).…”
