Establishment of human iPSC line NCCSi003-A from CD34 + cells of peripheral blood collected during apheresis of healthy donor from Indian ethnicity

Abstract: We present generation of iPSCs from CD34 cells isolated from peripheral blood, collected during apheresis of a healthy female individual. We nucleofected the CD34cells by episomal vectors containing Oct4, Sox2, L-Myc, Lin28, Klf4 and p53DD (dominant negative mutation in p53). The resultant colonies showed cobble-stone appearance and stained positive for alkaline phosphatase. The colonies demonstrated presence of pluripotency markers by immunofluorescence, flow-cytometry and PCR. The plasmids were lost from cel… Show more

“…The cascade filtration, and extracorporeal photopheresis, were reported only from one blood centre each. The centres from India are increasingly reporting the use of apheresis for ABO‐incompatible solid organ transplant, 17–20 therapeutic cytoreduction, 21,22 peripheral blood stem cell (PBSC) harvest, 23 and regenerative medicine for the establishment of human‐induced pluripotent stem cell lines 24 …”

“…In our lab, while reprogramming CD34 + cells from CB/apheresis sample, a few iPSC lines that displayed normal karyotype were established (Fernandes et al, ; Fernandes, Shinde, et al, ; Fernandes, Tembe, et al, ). During this process, one of the transfections resulted in development of a line that showed full trisomy of Chromosome 1 and a deletion of small fragment q23 to qter of Chromosome 11.…”

“…We have developed a few induced pluripotent stem cell (iPSCs) lines from cord blood (CB)/apheresis‐derived CD34 + cells of Indian ethnicity, using episomal vectors (Fernandes et al, ; Fernandes, Shinde, et al, ; Fernandes, Tembe, et al, ). While generating such lines, in one of the lines, we found an abnormality in Chromosomes 1 and 11.…”
