Establishment of a murine model of malignant mesothelioma

Abstract: Malignant mesothelioma (MM) is an aggressive tumour of the serosal cavities which is associated with previous asbestos exposure and is generally found to be resistant to conventional forms of therapy. Adequate scientific and clinical assessment of this disease has been severely limited by the relatively low incidence of mesothelioma and the lack of representative cell lines and animal models. The purpose of this study was to develop an asbestos-induced murine model of MM both as an in vivo-passaged malignancy … Show more

“…Murine mesothelioma cell lines have been derived for several mouse strains from primary asbestos induced tumours [17,18]. Intra-tumoural Treg cells in the C57BL/6J model of AE17 mesothelioma were found to increase significantly as a percentage of total CD4 + T cells within tumours as they grew [11].…”

“…20 Cells were cultured in ''R10'' medium that consisted of RPMI-1640 (Gibco /BRL, Grand Island, NY ) supplemented with 10% fetal calf serum (FCS ) ( CSL, Melbourne, Australia ) , gentamicin 48 mg / L (Sigma Chemical, St. Louis, MO ) , benzylpenicillin 60 mg / L (CSL ), 2 g/L sodium hydrogen carbonate (BDH, Victoria, Australia ), 20 mM N -2-hydro -2 -ethylpiperazine -N -2-ethane sulfonic acid ( HEPES ) ( Gibco /BRL ), and 2-mercaptoethanol (Sigma) . All cell lines used were tested regularly for mycoplasma contamination and were consistently found to be mycoplasma negative.…”
