Establishment of a Human Multiple Myeloma Xenograft Model in the Chicken to Study Tumor Growth, Invasion and Angiogenesis

Abstract: Multiple myeloma (MM), a malignant plasma cell disease, remains incurable and novel drugs are required to improve the prognosis of patients. Due to the lack of the bone microenvironment and auto/paracrine growth factors human MM cells are difficult to cultivate. Therefore, there is an urgent need to establish proper in vitro and in vivo culture systems to study the action of novel therapeutics on human MM cells. Here we present a model to grow human multiple myeloma cells in a complex 3D environment in vitro a… Show more

“…Instead of a mouse model, we here chose a chick chorioallantoic membrane (CAM) assay modified for the use with human myeloma cells as previously reported [26, 27]. This model has also been successfully utilized to investigate tumor-stroma interactions in other human B-cell malignancies [28, 29].…”

“…Myeloma cell lines (OPM-2 TetR and U266 TetR ) were generated as described (18). OPM-2 TetR and U266 TetR lines were further lentivirally transfected (pLenti6.3/GRP78-FLAG), and stable cell lines were selected over several weeks with 2.5 µg/ml blasticidin (Invitrogen).…”

“…Martowicz et al [ 67 ] used chicken embryos, as these are suitable for xenotransplantation due to a lack of adaptive immune responses until hatching [ 72 ]. The model combined the advantage of a 3D culture system with ex vivo development of chicken embryos.…”
