Establishment and Characterization of Canine Rhabdomyosarcoma Cell Line CMS-C

Abstract: ABSTRACT. A novel canine tumor cell line designated as the CMS-C cell line was established from pleomorphic rhabdomyosarcoma (RMS) raised in the prostate gland of a 14-year-old intact male mixed-breed dog. CMS-C cells displayed the same immunohistochemical characteristics (positive for vimentin and desmin and negative for cytokeratin and smooth muscle actin) as the original tumor cells and express myoD1 and UCP3, known as striated muscle-specific molecules, as shown by RT-PCR assay. Therefore, the established … Show more

“…RMS has a moderate metastatic rate, but can spread to the lungs, liver, spleen, kidneys and adrenal glands. Despite low metastatic rate, RMS is locally invasive (Azakami and others 2011). In agreement with the literature (Meuten 2002), we built Table 1 that summarises the main characteristics of the RM and RMS.…”

“…31 Pleomorphic RMS occurs mostly in adult people, 19,109 and the 2 cases in dogs have been in a 3-year-old and a 14-year-old dog. 8,31 Attempts have been made to segregate neoplasms into different categories for the purpose of making predictions of biologic behavior based on histomorphologic, ultrastructural, or immunohistochemical features. The most recent, and widely used, human classification system is the international classification system developed by Newton et al; 104 however, similar yet differing systems exist and may be encountered periodically in literature searches.…”

“…Although a tumor cell line was established from a pleomorphic RMS from a 14-year-old mixed breed dog, no cytogenetic abnormalities were reported. 8 One report described single or multiple RMS developing in 25 of 100 piglets from multiple litters over a 6-month period in the Netherlands, suggesting an inherited component. 157 van der Loop and colleagues established a cell culture from a pleomorphic RMS in a 3.5-week-old female piglet and described a deletion of the long arm of the X chromosome (Xq24-qter) in these cells.…”

“…The canine rhabdomyosarcoma cell line CMS-C [ 2 ] was maintained in Dulbecco’s modified eagle medium (DMEM, Wako, Osaka, Japan) with 10% heat-inactivated fetal calf serum (Hyclone, Logan, UT, U.S.A.) and antibiotics (Nakarai tesque, Kyoto, Japan) at 37°C in an atmosphere containing 5% CO 2 . CMS-J, bladder rhabdomyosarcoma from a 14-year-old spayed female beagle dog, was established as reported previously [ 2 ] and maintained in the same medium.…”
