Establishment and characterization of a Kaposi's sarcoma‐associated herpesvirus‐ and Epstein–Barr virus‐negative malignant lymphoma cell line (OHK) with primary effusion lymphoma immunophenotype

Abstract: Summary. A novel cell line, designated OHK, was established from ascites of a 59-year-old Japanese woman with diffuse large B-cell lymphoma showing a peculiar serosal tropism, as seen in primary effusion lymphomas (PEL). OHK exhibited a large pleomorphic morphology with irregular nuclei and distinct nucleoli, and included immunoblastic and Reed±Sternberg-like giant cells. On ultrastructural examination, rich intermediate ®laments, and well-developed Golgi apparati and rough endoplasmic reticulum, were seen. Im… Show more

“…We investigated VEGF and NP ‐1 expression in 21 hematopoietic cell lines, including OHK (a B cell lymphoma cell line with PEL immunophenotype 9 ). In reverse transcription–polymerase chain reaction (RT–PCR), the primers for VEGF and NP‐1 were designated according to Cornali et al 10 .…”

“…One angiogenetic factor involved in these mechanisms seems to be vascular endothelial growth factor (VEGF). Indeed, PEL cell lines, including OHK, produce high levels of VEGF (Aoki & Tosato, 1999; Kuwabara et al, 2002). Another factor may be HA; its involvement in angiogenesis was first presented in an excised chick chorio‐allantoic membrane, showing a positive angiogenic response to HA fragments (West et al , 1985).…”

“…The PEL cell line (OHK) was used in this study (Kuwabara et al, 2002). In addition, Jurkat (T‐cell acute lymphocytic leukaemia) and CA46 (Burkitt's lymphoma) cell lines were purchased from the American Type Culture Collection (Rockville, MD, USA).…”

“…Cultured malignant lymphoma cells [5] were metabolically labeled for 24 h with 10 µCi/ml of d -[6- 3 H]glucosamine (32 Ci/mol) in RPMI-1640 medium supplemented with 10% (v/v) heat-inactivated fetal bovine serum. The labeled materials in conditioned medium were precipitated at 0°C by the addition of 3 vol of 95% (v/v) ethanol containing 1.3% (w/v) potassium acetate, and then the precipitate was treated at room temperature for 24 h with 0.2 m NaOH followed by Pronase digestion (0.2 mg /ml of medium) for 24 h at 50°C.…”
