Establishment and characterisation of a new tumorigenic cell line with a normal karyotype derived from a human breast adenocarcinoma

Abstract: In vitro models are required for the study of these cancers, and several cell lines have already been established and characterised (Lafargues & Ozzello, 1958;Soule et al., 1973;Cailleau et al., 1974;Engel et al., 1978;Whitehead et al., 1983;Yamane et al., 1984;Chu et al., 1985;Vandewalle et al., 1987). Nevertheless, owing to the heterogenity and the diversity of mammary cancers, a great number of cell models is necessary to understand the reasons for this diversity and the effect of anticancer drugs on tumour… Show more

“…CAL51 was perfectly diploid, as previously reported by classical banding (Gioanni et al, 1990). MT-3 showed minimal deviation from diploidy -iso13 q, trisomy 7 and loss of X.…”

“…MCF 10A (ATCC-CRL 10317) were grown in DMEM Hams F12, 5% horse serum, hydrocortisone (0.5 mg/ml), cholera toxin (0.1 mg/ml), insulin (5 mg/ml) and EGF (0.01 mg/ml). (Whitehead et al, 1983) and Cal51 (Gioanni et al, 1990) were routinely cultured in DMEM, 10% FBS.…”

“…The chfr-specific probe (623 pb) was generated by PCR using 5 0 -AGT CCA GGA TTA CGT GTG CCC-3 0 (sense) and 5 0 -CCT CCA GTG CTG CTG AAA GCT GCT C-3 0 (antisense) primers mutation leading to MIN (Wheeler et al, 1999;Gayet et al, 2001). All breast cancer cell lines were aneuploid with the exception of HME-1 and Cal-51 (Gioanni et al, 1990;Forozan et al, 2000;Kytola et al, 2000;Rummukainen et al, 2001).…”
