Establishing Rps6 hemizygous mice as a model for studying how ribosomal protein haploinsufficiency impairs erythropoiesis

Abstract: Diamond-Blackfan Anemia(DBA) is a congenital hypoproliferative macrocytic anemia; 5q-syndrome myelodysplastic syndrome(MDS) is an acquired hypoproliferative macrocytic anemia. Their common erythroid phenotype reflects a shared pathophysiology -- haploinsufficiency of one of many ribosomal proteins and somatic deletion of one allele of the ribosomal protein S14 gene, respectively. Although these abnormalities lead to defective ribosome biogenesis, why ribosomal protein hemizygosity results in anemia is not cert… Show more

“…Decreased levels of granulocytes in the Sbds R126T/– model were also determined by flow cytometry of fetal liver cells (E16.5, S4A Fig ). Unlike ribosome deficiency models with dominant inheritance [ 52 54 ], erythrocyte levels prior to birth (E18.5) were normal in both mouse models ( S4B Fig ), consistent with observations in SDS patients [ 31 ].…”

“…Hematologic parameters are normal in these mice [50]. Also, unlike deletion of RPL22 in yeast [51], Rpl22 −/− mice have no substantial difference in growth rate or size relative to Rpl22 +/+ and Rpl22 +/− littermates (Anderson et al 2007 and our unpublished data).…”

“…There are several murine models of DBA (39, 40), but unfortunately, the hematopoietic phenotypes incompletely capture DBA. For example, mice heterozygous for Rps19 have normal erythropoiesis, even when stressed, and mice lacking Rps19 die before birth.…”
