Essential Role for Humoral Immunity duringEhrlichiaInfection in Immunocompetent Mice

Abstract: Although cellular immunity is essential for host defense during intracellular bacterial infections, humoral immunity can also play a significant role in host defense during infection by some intracellular bacteria, including the ehrlichiae. Antibodies can protect susceptible SCID mice from fatal Ehrlichia chaffeensis infection, an observation that has been hypothesized to involve the opsonization of bacteria released from host cells. To determine whether humoral immunity plays an essential role during ehrlichi… Show more

“…Passive transfer of anti-E. muris polyclonal antibodies completely protects SCID/B6 mice against lethal challenge with E. muris, and the protection is Fc dependent (5). Furthermore, mice deficient in B cells or Fc␥RI fail to resolve ordinarily sublethal IOE infection, suggesting a role for antibodies in protective immunity (40). Lack of a strong IgG antibody response in IOE infection is consistent with the observations reported in the previous studies (12,40).…”

Lack of a strong IgG antibody response in IOE infection is consistent with the observations reported in the previous studies (12,40). IgG antibodies are likely to confer protection against Ehrlichia by opsonization of host-cell-free bacteria and facilitation of their phagocytosis and killing (12).…”

“…Using one such agent, Ixodes ovatus ehrlichia (IOE), 3 we previously demonstrated that CD4 ϩ T cell-mediated IFN-␥-dependent production is critical for host defenses (19). Other studies from our laboratory support an essential role for Abs during IOE infection, a requirement that is not typical of intracellular bacterial infections (20). In contrast, CD8 T cells were not essential for protection against a low-dose IOE infection (19).…”

“…Recently, a number of studies demonstrated that humoral immunity plays an essential role in host defenses against ehrlichial pathogens (10,33,34,36). Furthermore, a small subset of E. chaffeensis proteins, many of which contain tandem repeats (TRs), appear to be the primary targets of the humoral immune response and are considered to be the major immunoreactive proteins (4,5,15,30).…”
