Escherichia coli stress response systems and their reaction to terahertz radiation

Abstract

In this review, we summarize the latest data concern ing the reactions of Escherichia coli to nonthermal terahertz radiation and the underlying molecular mechanisms. E. coli is the most simple and convenient model object for studying the effects of terahertz radiation: both its genetics and metabolism are well studied, and it is easily amenable to genetic engineer ing allowing one to create biosensors using promoters of genes activated by certain stress factors and the re porter GFP protein. Transformed E. coli cells containing biosensors can be used to visualize their reactions to terahertz radiation based on the intensity of GFP fluorescence. In this review, we present data on the response of certain E. сoli stress response sys tems to terahertz radiation obtained by us, as well as by other authors. We discuss experimental results for E. сoli/ pKatGGFP, E. сoli/pCopAGFP, and E. сoli/ pEmrRGFP biosensors that are used to detect E. сoli genetic net works responding to oxidative stress, copper ion ho meostasis failures, and antiseptics, res pectively. The obtained data indicate that exposure to nonthermal terahertz radiation induces E. сoli gene networks of oxidative stress and copper ion homeostasis, but does not activate those responding to antibiotics, protonophores, or superoxide anions. The fact that E. сoli/pKatGGFP and E. сoli/pCopAGFP biosensors have different activation and reaction periods when exposed to terahertz radiation and natural inducers suggests that reactions of oxidative stress and copper ion homeostasis systems to tera hertz radiation are specific.Key words: terahertz radiation; E. сoli stress response systems; biosensors.Изложены результаты последних лет по изу чению реакции стрес совых систем Escherichia coli в ответ на нетер мическое воздейст вие терагерцового (ТГц) излучения. Наиболее простым и удобным объектом для изучения воздействия ТГц излу чения на живые объекты является бактерия E. сoli. Это связано с ее высокой изу ченностью как на молекулярногенетическом, так и на метабо лическом уровне, а также c возможностью создания на основе промоторов ее стрессактивируемых генов и репор тер ного белка GFP геносенсорных конструкций. Введение этих кон струкций в клетки E. coli позволяет исследовать реакцию кон крет ной стрес совой системы бактерии на ТГц излучение по ин тен сивно сти син теза белка GFP, легко определяемого флуорометри чески. В рабо те представлены данные литературных источников и собственные результаты по нетермическому воздействию ТГц излучения на кон кретные стрессовые системы E. coli. Обсуждают ся эксперимен тальные данные, полученные с использованием геносенсоров E. coli/pKatGGFP, E. coli/pCopAGFP и E. coli/pEmrRGFP, которые являются маркерами генных сетей E. coli, активирующихся в усло виях окислительного стресса, при нарушении гомеостаза ионов меди и в присутствии антисептиков соответственно.
