Animals have evolved foraging strategies to acquire blends of nutrients that maximize fitness traits. In social insects, nutrient regulation is complicated by the fact that few individuals, the foragers, must address the divergent nutritional needs of all colony members simultaneously, including other workers, the reproductives, and the brood. Here we used 3D nutritional geometry design to examine how bumblebee workers regulate their collection of 3 major macronutrients in the presence and absence of brood. We provided small colonies artificial nectars (liquid diets) and pollens (solid diets) varying in their compositions of proteins, lipids, and carbohydrates during 2 weeks. Colonies given a choice between nutritionally complementary diets self-selected foods to reach a target ratio of 71% proteins, 6% carbohydrates, and 23% lipids, irrespective of the presence of brood. When confined to a single nutritionally imbalanced solid diet, colonies without brood regulated lipid collection and over-collected protein relative to this target ratio, whereas colonies with brood regulated both lipid and protein collection. This brood effect on the regulation of nutrient collection by workers suggests that protein levels are critical for larval development. Our results highlight the importance of considering bee nutrition as a multidimensional phenomenon to better assess the effects of environmental impoverishment and malnutrition on population declines.

The sexes perform different reproductive roles and have evolved sometimes strikingly different phenotypes. One focal point of adaptive divergence occurs in the context of diet and metabolism, and males and females of a range of species have been shown to require different nutrients to maximise their fitness. Biochemical analyses in 15Drosophila melanogaster have confirmed that dimorphism in dietary requirements is associated with molecular sex-differences in metabolite titres. In addition, they also showed significant within-sex genetic variation in the metabolome. To date however, it is unknown whether this metabolic variation translates into differences in reproductive fitness. The answer to this question is crucial to establish whether 20 genetic variation is selectively neutral or indicative of constraints on sex-specific physiological adaptation and optimisation. Here we assay genetic variation in consumption and metabolic fitness effects by screening male and female fitness of thirty D. melanogaster genotypes across four protein-to-carbohydrate ratios. In addition to confirming sexual dimorphism in consumption and fitness, we find 25 significant genetic variation in male and female dietary requirements. Importantly, these differences are not explained by feeding responses and most likely reflect metabolic variation that, in turn, suggest the presence of genetic constraints on metabolic dimorphism. 30

Symbiotic bacteria support host growth upon malnutrition. How bacteria achieve this remains partly elusive. Here, we took advantage of the mutualism between Drosophila and Lactiplantibacillus plantarum (Lp) to investigate such mechanisms. Using chemically-defined holidic diets, we found that association with Lp improves the growth of larvae fed amino acid-imbalanced diets. We show that in this context Lp supports its host’s growth through a molecular dialog that requires functional operons encoding ribosomal and transfer RNAs (r/tRNAs) in Lp and the GCN2 kinase in Drosophila’s enterocytes. Our data indicate that Lp’s r/tRNAs loci products activate GCN2 in a subset of larval enterocytes, a mechanism necessary for the host’s adaptation to amino acid imbalance that ultimately supports growth. Our findings unravel a novel beneficial molecular dialog between hosts and microbes, which relies on a non-canonical role of GCN2 as a mediator of non-nutritional symbiotic cues encoded by r/tRNA operons.
