ERK1/2 inhibition promotes robust myotube growth via CaMKII activation resulting in myoblast-to-myotube fusion

Abstract: Summary Myoblast fusion is essential for muscle development and regeneration. Yet, it remains poorly understood how mononucleated myoblasts fuse with preexisting fibers. We demonstrate that ERK1/2 inhibition (ERKi) induces robust differentiation and fusion of primary mouse myoblasts through a linear pathway involving RXR, ryanodine receptors, and calcium-dependent activation of CaMKII in nascent myotubes. CaMKII activation results in myotube growth via fusion with mononucleated myoblasts at a fusoge… Show more

“…However, after examining in detail the expression of which genes are downregulated under short-term HS, we found that the expression of gene clusters for BMP-signaling, ERK1/2-signaling, and pathways involved in response to TGF-β is suppressed ( Table 1 ). According to literature data, suppression of the activity of these signaling pathways leads to premature myogenic differentiation [ 32 , 33 ]. At the early stages of SCs activation, BMP signaling pathway promotes an increase of the pool of proliferating cells and prevents premature differentiation in myoblasts [ 33 ].…”

“…It is also known that a decrease in the activity of the ERK1/2 signaling pathway leads to a reduction in the proliferation rate of myoblasts [ 34 ]. In addition, it has been shown that inhibition of the ERK1/2 signaling pathway leads to the activation of calcium-dependent mechanisms causing robust differentiation of activated SCs [ 32 ]. In the present study, we observed a decrease in the expression of genes of the ERK1/2 pathway in myoblasts isolated from the soleus muscle after 1 day of HS ( Table 1 ).…”
