ER stress regulating protein phosphatase 2A-B56γ, targeted by hepatitis B virus X protein, induces cell cycle arrest and apoptosis of hepatocytes

Abstract: Hepatitis B virus X (HBx) protein contributes to the progression of hepatitis B virus (HBV)-related hepatic injury and diseases, but the exact mechanism remains unclear. Protein phosphatase 2 A (PP2A) is a major serine/threonine phosphatase involved in regulating many cellular phosphorylation signals that are important for regulation of cell cycle and apoptosis. Does HBx target to PP2A-B56γ and therefore affect HBx-induced hepatotoxicity? In the present study, the expression of B56γ positively correlated with … Show more

“…During productive virus infection, viruses produce a large amount of viral proteins accumulated in the ER lumen (13)(14)(15). Some viruses, such as hepatitis C virus (HCV) and dengue virus (DENV), also use the ER as their replication sites (16,17).…”

“…HepaRG cells were obtained from the National Institute of Diagnostics and Vaccine Development in Infectious Diseases (Xiamen University). Cell culture and differentiation for HepaRG cells were described in our previous study (He et al 2018). Human normal hepatic cell line L02 and human embryonic kidney cell line HEK-293T were maintained in our laboratory, cultured in RPMI 1640 medium (Hyclone, UT, USA) and in Dulbecco's Modified Eagle Medium (DMEM) high glucose medium (Hyclone) which was supplemented with 10% FBS and 1% (v/v) penicillin-streptomycin, respectively.…”

“…Once the molecular adaption fails in resolving the proteinfolding defect, hepatocytes enter persistent ER stress, which results in apoptosis 5 . ER stress-related apoptosis has been found in fatty liver disease, viral hepatitis, and alcohol or drug-induced liver injury 3,6,7 . The transcription factor C/EBP homologous protein (CHOP) mediates the most well-characterized proapoptotic pathway resulted from unresolved ER stress.…”
