Equivocal Association of RAD51 Polymorphisms with Risk of Esophageal Squamous Cell Carcinoma in a Chinese Population

Abstract: Aim: To study the contribution of genetic variation in RAD51 to risk of esophageal squamous cell carcinoma (ESCC). Methods: Three single nucleotide polymorphisms (SNPs) in RAD51 (rs1801320, rs4144242 and rs4417527) were genotyped in 316 ESCC patients and 316 healthy controls in Anyang area of China using PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism). Demographic variables between cases and controls were statistically compared by T test and Chi-square test. Hardy-Weinberg equilib… Show more

“…To the best of our knowledge, no previous studies have evaluated this SNP in oral and oropharyngeal SCCs separately, and the results for head and neck SCC are contradictory. No association between RAD51 rs1801320 and esophageal SCC susceptibility in the Chinese population (Zhang, Yang, et al, ) and head and neck SCC occurrence in the Polish population (Gresner et al, ) was reported, but RAD51 rs1801320 was a significant susceptibility factor in laryngeal SCC when associated with smoking and drinking (Romanowicz‐Makowska et al, ). On the other hand, the minor allele of RAD51 rs1801320 was associated with a significantly reduced risk for head and neck SCC in populations from Belgium (Werbrouck et al, ) and Poland (Sliwinski et al, ).…”

“…Meanwhile, genetic variant in RAD51 was investigated susceptibility of ESCC. the haplotype of three SNPs in RAD51 increased the ESCC risks (Zhang et al, 2014). PLEC1 on 10q23, C20orf54 on chromosome 20p13, SLC39A6 on chromosome 18q22 was identified by GWAS to be significantly associated with susceptibility to ESCC in Chinese population (Abnet et al, 2010;Wang et al, 2010;Wu et al, 2011a).…”

“…However, the results are still inconsistent. For instance, Sun et al [39] demonstrated G135C polymorphism in RAD51 gene was strongly related to EC, while Zhang et al [37] did not discover the significant relationship between EC and polymorphisms in RAD51. So the meta-analysis was conducted to examine the correlation between the polymorphisms in RAD51 gene and the susceptibility to HNC and EC.…”
