Epitope‐Resolved Detection of Peanut‐Specific IgE Antibodies by Surface Plasmon Resonance Imaging

Abstract: Peanut allergy can be life-threatening and is mediated by allergen-specific immunoglobulin E (IgE) antibodies. Investigation of IgE antibody binding to allergenic epitopes can identify specific interactions underlying the allergic response. Here, we report a surface plasmon resonance imaging (SPRi) immunoassay for differentiating IgE antibodies by epitope-resolved detection. IgE antibodies were first captured by magnetic beads bearing IgE ε-chain-specific antibodies and then introduced into an SPRi array immob… Show more

“…SPRi-based biosensors for detecting SARS-CoV-2 have also been developed. These sensors follow the principles of SPR and have been utilised to study binding kinetics between biomolecular species in the past [ 52 , 53 , 54 ]. SPRi assay was used for quantitative measurement of IgG, IgM and IgA antibodies binding to the RBD spike protein in serum sample [ 43 ].…”

“…Carboxylated magnetic beads (MB, 1.5 μm diam.) were used to immobilize IgGs since they are very effective for analyte capture and isolation. IgGs were oriented through protein A/G-mediated Fc binding, and amine-to-carboxyl IgG conjugation was used to achieve randomly oriented controls (Scheme ). These two successful immobilization strategies are well accepted and thus used in this report as different immobilization models.…”

“…The results indicate that the MAG–protein A/G bead strategy for isolating GADA from serum reduced the nonspecific background signals from proteins and other components present in the free-serum matrix and thus offered enhanced sensitivity (Figure A). , The simplicity of the magnetic capturing and isolation of bound GADA from free serum by the MAG–protein A/G beads is not feasible with other nonmagnetic nanomaterials, which would require centrifugation or other tedious separation procedures.…”
