Epitope Diversity of N-Glycans from Bovine Peripheral Myelin Glycoprotein P0 Revealed by Mass Spectrometry and Nano Probe Magic Angle Spinning 1H NMR Spectroscopy

Abstract: The carbohydrate structures present on the glycoproteins in the central and peripheral nerve systems are essential in many cell adhesion processes. The P0 glycoprotein, expressed by myelinating Schwann cells, plays an important role during the formation and maintenance of myelin, and it is the most abundant constituent of myelin. Using monoclonal antibodies, the homophilic binding of the P0 glycoprotein was shown to be mediated via the human natural keller cell (HNK)-1 epitope (3-O-SO 3 H-GlcUA(␤1-3)Gal(␤1-4)G… Show more

“…The method has also been used to detect nanomole amounts of purified lipopolysaccharide (54), purified O-linked and N-linked glycopeptides (8,9,56,57), and LOS ganglioside mimics (11). In this study, HR-MAS NMR was used to further examine CPS directly from campylobacter cells.…”

“…By means of mass spectrometry and HR-MAS nano-NMR techniques (11,37), the structure of the Asn-linked glycan was established as a heptasaccharide comprising a linear chain of one D-Bac and five D-GalNAc residues with a D-Glc branch (Fig. 6a).…”

“…The HNK-1 carbohydrate structure on P0 and the neural cell adhesion molecule, the major carrier glycoproteins in the peripheral and central nervous systems, respectively, has been determined (27)(28)(29). All of the HNK-1 carbohydrate structure determined was of the sulfated form, i.e.…”
