Epithelial mouse mammary cell line exhibiting normal morphogenesis in vivo and functional differentiation in vitro.

Abstract: An epithelial cell line, designated COMMA-1D, was derived from mammary tissue of BALB/c mice in the middle of pregnancy. This Considerable effort has been directed toward the characterization of normal, preneoplastic, and neoplastic mammary cells and tissues in vivo and in vitro with respect to growth regulation, hormonal responsiveness, and biochemical properties (for a review, see ref.2). As part of this effort, a significant amount of work has been directed toward the development of cell culture methodolo… Show more

“…ORAI1-mediated STIM2-activated enhancement of basal calcium influx during lactation would be an effective way for mammary gland epithelial cells to ensure Ca 2þ was available to enrich milk with Ca 2þ . To assess the potential role of Stim2 in mammary epithelial cells during lactation, we used the HC11 mouse mammary epithelial cell line model (36). Differentiation of HC11 cells using lactogenic hormones significantly induced the mRNA expression of the milk protein b-casein (data not shown), which was undetectable prior to differentiation (16,37).…”

“…The first spontaneously immortalized cell line pertinent to breast biology, COMMA-1D, was isolated from normal mid-pregnant BALB/c mice (Danielson et al 1984). In contrast to mouse cells, nonmalignant human cells do not immortalize spontaneously (Harris 1987).…”

“…HC11 mammary epithelial cells, 19) which had been cloned as a casein-expressing subline from the COMMA-1D mouse mammary epithelial cell line, which itself had been established from a mouse mammary epithelial primary culture, 20) were kindly provided by Dr. B. Groner (Georg Speyer Haus, Institute for Biomedical Research, Frankfurt am Main, Germany). The HC11 cells were maintained in Dulbecco's modified Eagle's medium (DMEM, 4,500 mg/L of glucose, Sigma) containing 10% fetal calf serum (FCS) and 10 ng/mL of EGF (Sigma).…”
