Epithelial lining fluid free IGF-I-to-PAPP-A ratio is associated with bronchopulmonary dysplasia in preterm infants

Abstract: Preterm newborns developing retinopathy of prematurity (ROP) and bronchopulmonary dysplasia (BPD) show persistently low levels of insulin-like growth factor-I (IGF-I) in sera. They also present higher free IGF-I concentrations in epithelial lining fluids (ELFs) and lung tissues. Pregnancy-associated plasma protein-A (PAPP-A) is a metalloproteinase that dissociates three binding proteins from the active form of IGF-I, namely free IGF-I. The present study analyzes the ELF concentrations of free IGF-I, PAPP-A, an… Show more

“…Proteins and urea were measured, as previously described [22,23]. Serum urea values obtained during the routine clinical tests in the same day of the lavage were used to calculate the serum-to-BALF urea ratio and obtain epithelial lining fluid (ELF) concentrations.…”

“…Urea was measured using a high-sensitivity colorimetric method on an automatic analyser, as previously described [25]. All laboratory assays were performed in duplicate, by investigators blind to the clinical data.…”

“…Data were corrected for the serum-to-BAL urea ratio in order to transform BAL into epithelial lining fluid (ELF) concentrations [25]. Measurement results in meconial solutions were compared as raw data or as per gram of meconium (after correction for the dilution).…”

“…The serum/BAL urea ratio was used to correct for dilution and obtain epithelial lining fluid (ELF) concentrations [12]. Surfactant function in BAL was studied via fluorescence analysis of adsorption kinetics into the air/liquid interface, as described previously [13].…”
