Epigenetics of Male Infertility: The Role of DNA Methylation

Abstract: In recent years, a number of studies focused on the role of epigenetics, including DNA methylation, in spermatogenesis and male infertility. We aimed to provide an overview of the knowledge concerning the gene and genome methylation and its regulation during spermatogenesis, specifically in the context of male infertility etiopathogenesis. Overall, the findings support the hypothesis that sperm DNA methylation is associated with sperm alterations and infertility. Several genes have been found to be differentia… Show more

“…Adenosine binding to A 1 R/A 3 R causes a decrease in cAMP, while adenosine binding to A 2 AR and A 2 BR causes an increase in cAMP. Evidence indicates that ARs expression appears to be mediated by DNA methylation [ 16 ], which is a critical epigenetic process for gene expression regulation [ 17 , 18 ] in a variety of cell types [ 19 23 ].

“…In the respiratory tract, it is expressed in human epithelial alveolar cells, where it favors the SARS-CoV-2 infection [48]. Furthermore, similarly to other genes [50][51][52][53], the hACE2 gene is under epigenetic regulation by DNA methylation [54]. Recent findings indicate that its improper methylation might increase SARS-CoV-2 susceptibility [54].…”

“…This process predominantly occurs in CpG dinucleotides [ 71 ]. Regions rich in CpGs are typically proximal to regulatory regions, e.g., promoters [ 70 , 72 , 73 , 74 ], which regulate gene expression [ 75 ]. When methylated, these regions usually lead to gene silencing [ 70 ].…”
