Epigenetics in neurodegenerative disorders induced by pesticides

Abstract: Neurodegenerative diseases are becoming major socio-economic burdens. However, most of them still have no effective treatment. Growing evidence indicates excess exposure to pesticides are involved in the development of various forms of neurodegenerative and neurological diseases through trigger epigenetic changes and inducing disruption of the epigenome. This review summaries studies on epigenetics alterations in nervous systems in relation to different kinds of pesticides, highlighting potential mechanism in … Show more

“…DNA methylation possesses a critical role in maintaining cognitive functions through gene regulation and other mechanisms in the CNS . DNMTs, such as DNMT1, DNMT3A, and DNMT3B, catalyze DNA methylation by transferring methyl groups from the methyl donor SAM to certain DNA bases .…”

“…27 According to these studies, our findings showed that the LPS-treated adverse effects, such as the elevated levels DNA methylation possesses a critical role in maintaining cognitive functions through gene regulation and other mechanisms in the CNS. 30 DNMTs, such as DNMT1, DNMT3A, and DNMT3B, catalyze DNA methylation by transferring methyl groups from the methyl donor SAM to certain DNA bases. 31 While DNMT3A and DNMT3B are primarily in charge of producing new methylation patterns, DNMT1 is primarily in charge of maintaining current methylation patterns.…”

“…The nucleosome consists of 147 bp of DNA wrapped around an octamer of histones composed of two H3/H4 dimers and two H2A/H2B dimers [ 33 ]. The dynamic and reversible modifications of histone proteins influence all DNA-based processes, including DNA packaging and chromatin compaction, nucleosome dynamics, and transcription [ 76 , 77 ]. This process is regulated by several post-translational modifications, most of which occur within the amino terminal in histone tails [ 78 ].…”

“…Both epidemiologic and experimental studies have evidenced that pesticides exposure affects central nervous systems, probably via epigenetic modifications ( Yu et al, 2021 ), such as up-regulation of miRNA-181 that drives the neuronal cells apoptosis as observed by Zhao et al (2019) in a neuroblastoma cell line. miRNA-181 up-regulation leads to increased oxidative stress, altered neurotransmitter status, neuroinflammation, neurodegeneration, and neuronal apoptosis.…”
