Epigenetics and Testicular Cancer: Bridging the Gap Between Fundamental Biology and Patient Care

Abstract: Testicular cancer is the most common solid tumor affecting young males. Most testicular cancers are testicular germ cell tumors (TGCTs), which are divided into seminomas (SGCTs) and non-seminomatous testicular germ cell tumors (NSGCTs). During their development, primordial germ cells (PGCs) undergo epigenetic modifications and any disturbances in their pattern might lead to cancer development. The present study provides a comprehensive review of the epigenetic mechanisms–DNA methylation, histone post-translati… Show more

“…Furthermore, young age should represent an element in favor of good tolerance to treatment, regardless of any known side effects related to CAR-T cell therapy ( 95 ). Furthermore, a better understanding of the mechanisms of activation of the immune system, including epigenetic influence in immune checkpoint expression ( 108 ), could also lead to a reconsideration of therapy with immune checkpoint inhibitors against GCTs.…”

“…Epigenetic modifications are increasingly recognised as important features of GCTs [33]. Hypermethylation and global remodelling of DNA methylation were reported to be key factors in mediating cisplatin resistance of testicular GCTs [34].…”

“…Similarly, KAT2B (PCAF) is overexpressed in TGCT and potentially promotes tumor progression by acetylating and activating the androgen receptor (AR). KAT6A and KAT9 are also overexpressed families in TGCT [96].…”

“…In addition, SET1A is responsible for the mono-, di-, and trimethylation of histone H3 at lysine 4 (H3K4), which is associated with active transcription. Thus, SET1A could be a potential therapeutic target for testicular cancers [96].…”
